接骨续筋口服液对家兔桡骨骨痂中OPG及破骨细胞凋亡率的影响

摘　　要：目的:观察接骨续筋口服液对家兔桡骨骨痂中骨保护素(Osteoprotegerin,OPG)产生及破骨细胞凋亡率的影响。方法:由安医大动物实验中心提供68只健康8个月龄的新西兰大白兔,雌雄皆用,根据该实验的要求采取随机数字表法随机分成五组,每组平均12只,考虑到死亡,预留8只备用,按照药物以及剂量的不同分为五组,造模术后分别予接骨续筋口服液低、中、高剂量、云南白药干预、空白对照模型组不予任何干预,分别造模术后1W、2W、4W采集标本,检验,获得数据,进行统计分析。结果:在骨折造模术后1W,2W,4W接骨续筋口服液高剂量组骨痂中OPG的数值分别为230.79±4.96、304.80±9.41、275.75±5.07,相同时间点与其他各组组间比较,免疫组化总数目高于其他各组(P<0.05);不同时间点组内比较2W高于1W、4W(P<0.01);高剂量组接骨续筋口服液破骨细胞凋亡率分别为23.58±1.43、59.19±4.073、14.19±1.55,相同时间点与其他各组组间比较,接骨续筋口服液高剂量组凋亡率明显高于其余各组,不同时间点组内比较2W高于1W、4W。结论:高剂量接骨续筋口服液在骨折愈合实验中具有明显升高骨痂中OPG的作用,并使破骨细胞使凋亡速度加快,减少骨折断端骨痂中破骨细胞数量,促进骨折愈合。Purpose:Observe the effect of Jieguxujing Oral Liquid on the production of OPG and the apoptosis rate of osteoclast in rabbits’radius poromas.Method:688-month-old New Zealand white rabbits,males and females,provided by the Animal Experimental Center of Anhui Medical University,were grouped according to experimental requirements.12 rabbits were randomly selected as items in the sham-operated group.8 rabbits were reserved as items in the accidental death group.The remaining 60 rabbits were selected at random and equally divided into 5 groups according to medicine and the dosage,including the low-dose group,the medium-dose group,the high-dose group,Yunnan Baiyao group,and the model group.Result:Statistical data show that rabbits took Jieguxujing Oral Liquid for 1 week,2 weeks and 4weeks after the fracture surgery,the immunohistochemical amounts of OPG in the high-dose group,which was detected at different time within the same group and at the same time among different groups,is higher than others.According to the statistic analysis of the apoptosis rate of osteoclast at different time within the same group and at the same time among different groups,the amounts of osteoclast in the high-dose group is the lowest and the apoptosis rate of osteoclast in the high-dose group is the highest.Conclusion:Jieguxujing Oral Liquid can accelerate the production of OPG and the apoptosis rate of osteoclast.Thus it can inactivate the osteoclast in broken poromas so as to regenerate the osteoblast and accelerate healing.
