摘  要:目的探讨红景天苷通过碱性成纤维细胞因子(bFGF)、血管内皮细胞生长因子(VEGF)抵抗成骨细胞凋亡的作用。方法选取MG-63人成骨细胞接种于96孔板进行培养,依据随机数字表分为红景组和对照组,每组48孔。对照组给予常规培养,红景组在此基础上给予100 nmol/L红景天苷培养。采用Western blot(WB)法检测bFGF、VEGF表达水平,采用细胞增殖实验(MTT)检测MG-63人成骨细胞增殖活性,采用骨化结节染色实验检测MG-63人成骨细胞骨化活性。结果红景组培养1、3 d bFGF、VEGF的WB值明显高于对照组,差异有统计学意义(P<0.05);红景组培养1、3 d MTT值(A492)明显高于对照组,差异有统计学意义(P<0.05);红景组培养1、3 d骨化结节数明显高于对照组,差异有统计学意义(P<0.05)。结论红景天苷可能通过上调bFGF、VEGF表达来抵抗MG-63人成骨细胞的凋亡,有助于改善MG-63人成骨细胞的骨化活性。Objective To investigate the effects of salidroside against osteoblast apoptosis through the basic fibroblast growth factor(bFGF)and vascular endothelial growth factor(VEGF).Methods MG-63 osteoblasts were cultured in 96-well plate.According to the random number table,which were divided into Hongjing group and control group,with 48 holes in each group.The osteoblasts in control group were cultured routinely,and the osteoblasts in Hongjing group,on the basis of control group,were treated by giving 100nmol/L salidroside.The Western blot(WB)was used to detect the expression levels of bFGF and VEGF,MTT was used to detect the proliferation activity of MG-63 human osteoblasts,moreover,the nodule ossification test was used to detect the ossification activity of MG-63 human osteoblasts.Results The WB values of bFGF and VEGF in Hongjing group were significantly higher than those in control group after 1-day and 3-day culture(P<0.05).The MTT values(A492)in Hongjing group were significantly higher than those in control group after 1-day and 3-day culture(P<0.05).Moreover the number of ossified nodules in Hongjing group was significantly higher than that in control group after 1-day and 3-day culture(P<0.05).Conclusion Salidroside may inhibit the apoptosis of MG-63 human osteoblasts by up-regulating the expression levels of bFGF and VEGF,which is helpful to improve the ossification activity of MG-63 human osteoblasts.
