摘 要:目的研究头低位卧床模拟失重30d及高+Gx暴露后猕猴腮腺组织腮腺分泌蛋白(PSP)表达的变化,探讨长时间失重及高+Gx暴露对腮腺分泌功能的影响。方法23只猕猴随机分为4组:正常对照组(A组,3只);模拟失重30d组(B组,3只);+13Gx/230s暴露组(C组,3只);模拟失重30d后高+Gx暴露组(D组,14只)。D组依据+Gx值又分为4个亚组,分别是+11Gx/270s暴露组(D1组,3只),+13Gx/230s暴露组(D2组,4只),+15Gx/200s暴露组(D3组,4只),+13Gx/230s暴露后恢复9d组(D4组,3只)。猕猴-10°头低位卧床建立失重模型,使用58型动物离心机建立高+Gx暴露模型。利用实时定量PCR(qRT-PCR)和蛋白印迹法(Western blot)检测猕猴腮腺组织PSP在mRNA及蛋白水平表达变化。结果与对照组相比,各实验组猕猴腮腺组织PSP蛋白和mRNA表达显著减少。失重30d后高+13Gx较单纯高+13Gx组相比,PSP表达显著减少(P<0.01)。模拟失重后再高+Gx暴露,猕猴腮腺组织PSP表达呈现随+Gx值增大而减弱的趋势,而恢复9d后表达回升。结论模拟失重30d及高+Gx暴露导致猕猴腮腺组织PSP表达降低,腮腺分泌免疫因子的功能受到影响。模拟失重降低了腮腺的超重耐受力,腮腺的损伤有可逆性。Objective To observe the changes of high+Gx exposure and 30dsimulated weightlessness on parotid secretory protein(PSP)expression,and to explore the effects of prolonged weightlessness and high+Gx exposure on the secretion function of parotid gland in rhesus macaques.Methods Twenty-three healthy male rhesus macaques were randomly divided into four groups:control group(group A,n=3);30dsimulated weightlessness group(group B,n=3);+13Gx/230sgroup(group C,n=3);high+Gx after 30dsimulated weightlessness exposure group(group D,n=14).Group D was further divided into four subgroups according to the+Gx load:+11Gx/270s(group D1,n=3),+13Gx/230s(group D2,n=4),+15Gx/200s(group D3,n=4),9drecovery after+13Gx/230s(group D4,n=3).The monkeys underwent a 30d-10°HDT bed-rest position to simulate weightlessness.Advanced 58-type animal centrifuge was used to establish the animal model of high+Gx load.The expression of PSP in rhesus macaque parotid glands on protein and mRNA level was separately detected by western blot and quantitative real-time PCR(qRT-PCR).Results Western blot analysis showed that PSP expression of rhesus macaque parotid glands in the experimental groups decreased significantly as compared with that in control group.The expression of PSP in 30dsimulated weightlessness and+13Gx/230sexposure group decreased significantly than that in simple+13Gx/230sgroup(P<0.01).In groups of high+Gx exposure after simulated weightlessness,PSP expression showed a decreasing trend with the increase of+Gx.However,it rose again in group D4after 9drecovery.The mRNA level displayed a conformably trend with that of the protein level.Conclusion The PSP expression of parotid gland in rhesus macaques decreases after 30dsimulated weightlessness and high+Gx exposure,and the function of secretion of immune factors by parotid gland is affected.Moreover,simulated weightlessness reduces the Gx tolerance of parotid gland,and the damage of parotid gland is reversible.
