摘  要:目的:探究一种稳定的、高效的建立巴马小型猪冠状动脉粥样硬化的模型。方法:将55只雄性巴马小型猪随机分为正常组(空白组)5只和手术组50只(含模型组7只),分别予普通饲料喂饲和高脂饲喂,2周后进行机械损伤术,共饲养48周,并分别于24、28、32和48周观察测定的相关指标。结果:计算机辅助的定量分析结果示24周正常组面积狭窄率为(4.48±0.98)%,模型组为(41.70±2.81)%,48周正常组面积狭窄率为(12.56±2.22)%,模型组为(67.06±5.67)%。冠状动脉血管内超声结果示24周正常组面积狭窄率为6.56%,模型组面积狭窄率为44.12%,48周正常组面积狭窄率为15.00%,模型组面积狭窄率为68.86%。血脂指标胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、氧化修饰低密度脂蛋白(ox-LDL)水平随时间延长,模型组均逐渐升高,高密度脂蛋白胆固醇(HDL-C)水平虽呈下降趋势,但仍高于正常组(P<0.01,P<0.05)。模型组炎性因子超敏C反应蛋白(hs-CRP)、白细胞介素-6(IL-6)、血管细胞黏附分子-1(VCAM-1)、单核细胞趋化蛋白-1(M C P-1)、P选择素(s P-s e l e c t i n)水平明显高于同期正常组且随时间延长而升高(P<0.05,P<0.01)。HE染色示模型组冠状动脉内膜损伤严重。结论:该方法可在短时间内成功建立巴马小型猪冠状动脉粥样硬化模型,为相关研究提供了可靠模型,并有利于进一步开展深入研究。Objective:To establish a stable and efficient model of coronary atherosclerosis in Bama miniature pig.Methods:Fifty-five male Bama miniature pig were randomly divided into blank group(n=5)and surgical group(n=50),which containing model group(n=7).The pigs in the blank group were fed with regular diet and the pigs in the surgical group were fed with high-fat diet.After 2 weeks,mechanical injury was performed,and 48 weeks were fed.The related indexes were observed at 24,28,32 and 48 weeks respectively.Results:QCA results showed that at week 24,stenosis ratio of blank group was(4.48±0.98)%and model group was(41.70±2.81)%.At week 48,area stenosis ratio of blank group was(12.56±2.22)%and model group was(67.06±5.67)%.IVUS results showed that at week 24,the stenosis ratio was 6.56%for the blank group and 44.12%for the model group.At week 48,the stenosis ratio was 15.00%for the blank group and 68.86%for the model group.TC,TG,LDL-C and ox-LDL levels of surgical group were higher than blank group(P<0.05,P<0.01).HDL-C level increased at first and then decreased.At the 24th,28th,36th and 48th week,hs-CRP,IL-6,VCAM-1,MCP-1 and sP-selectin levels of surgical group were higher than blank group(P<0.05,P<0.01).HE staining results showed that the coronary artery intima injury was serious of surgical group.Conclusion:The atherosclerosis induction by concomitant application of high fat diet and coronary mechanical abrasion is proved effective with satisfactory reliability and stability.
