摘　　要：目的:观察针灸十三鬼穴对抑郁大鼠外侧缰核βCaMKⅡ蛋白及其mRNA表达的影响,探讨十三鬼穴抗抑郁机制。方法:60只SD大鼠随机分正常组、模型组、针刺穴位组、针刺非穴组、艾灸穴位组、艾灸非穴组,每组10只。采用慢性轻度不可预见性应激法造模,针灸大陵、上星穴干预,定期进行体质量及行为学监测,应用Western Blot、qPCR检测外侧缰核βCaMKⅡ蛋白与mRNA表达。结果:与正常组比较,模型组大鼠体质量、糖水消耗、旷场分数均显著降低(P<0.05),外侧缰核中βCaMKⅡ蛋白与mRNA表达量显著上升(P<0.05);与模型组比较,针刺穴位组与艾灸穴位组均可有效逆转抑郁大鼠行为学的改变(P<0.05),下调βCaMKⅡ蛋白与mRNA的过度表达(P<0.05),且针刺穴位组效应更明显;各非穴组差异无统计学意义。结论:针灸十三鬼穴治疗抑郁有效,可能与下调抑郁模型大鼠外侧缰核中βCaMKⅡ蛋白及其mRNA的过度表达有关。Objective:To observe the effects of acupuncture and moxibustion of thirteen ghost points on the protein and mRNA expression ofβCaMKⅡin lateral habenular nucleus of depressive rats to discuss the anti-depression mechanisms of thirteen ghost points.Methods:Sixty male SD rats were randomly divided into 6 groups,the normal group,the model group,the acupoint group,the acu-non-point group,the moxibustion point group and the moxi-non-point group with 10 rats in each group.CUMS procedure was executed to establish depression model,and Daling(PC7)and Shangxing(DU23)from ghost points were treated by acupuncture and moxibustion respectively,body weight and behavior test were monitored regularly.Western Blot and quantitative polymerase chain reaction(qPCR)were used to detect the expression ofβCaMKⅡprotein and mRNA in lateral habenula.Results:Compared with the normal group,the body weight,source preference consumption,open field scores of the model rats were significantly decreased(P<0.05),and the expression ofβCaMKⅡprotein and mRNA in the lateral habenular nucleus was significantly increased(P<0.05);compared with the model group,acupoint and moxibustion point groups could effectively reverse the behavioral changes of depressive rats(P<0.05),and down-regulate the over-expression ofβCaMKⅡprotein and mRNA(P<0.05).The effect of acupoint group was more significant,while there was no significant difference among nonpoint groups.Conclusion:Acupuncture and moxibustion at thirdteen ghost points are effective in treating depression,the mechanism of which may be related to down-regulate the over-expression ofβCaMKⅡprotein and its mRNA in lateral habenular nucleus of depressive rats.
