补肾益气活血方对Stat3活化抑制大鼠胚胎着床及PR表达的影响

摘　　要：目的:通过宫腔注射信号传导与活化转录因子3(St a t3)抑制剂抑制大鼠胚胎着床,探讨补肾益气活血方改善胚胎着床的有关机制。方法:将妊娠雌鼠随机分为Stat3抑制剂组(SI组)及Stat3抑制剂加中药组(SI+BR组),并于妊娠第4天行宫腔注射(右侧:Stat3抑制剂;左侧:0.9%氯化钠溶液作为对照),SI+BR组灌胃补肾益气活血方,SI组则给予等体积无菌蒸馏水。收集妊娠第8天大鼠子宫标本,记录各组大鼠每侧子宫胚胎着床情况,免疫印迹法检测子宫内膜PR、Stat3及p-Stat3蛋白表达,RT-PCR检测子宫内膜PR mRNA表达。结果:SI组及SI+BR组与自身对照侧比较,抑制剂侧子宫着床胚胎数均显著减少(P<0.01),子宫内膜PR及p-Stat3/Stat3表达均显著下降(P<0.01),PR mRNA表达均显著降低(P<0.01)。两组各对照侧、抑制剂侧着床胚胎数、子宫内膜PR及p-Stat3/Stat3蛋白及PR mRNA表达比较,差异均无统计学意义。结论:补肾益气活血方改善大鼠胚胎着床的机制可能与其激活Stat3并上调PR表达,改善子宫内膜容受性有关。Objective:To explore the mechanism of Bushen Yiqi Huoxue Formula(BR)in improving embryo implantation by using intrauterine injection of signal transducers and activators of transcription 3(Stat3)inhibitor.Methods:The pregnancy rats were randomly allocated into group of Stat3 inhibitors(SI)and Stat3 inhibitors plus Chinese medicine group(SI+BR).On the day 4 of pregnancy,each rat was given intrauterine injection of a Stat3 inhibitor into the uterine lumen on the right side,while the left side was served as control by injection saline.The SI+BR group was orally given BR,while the SI group was given equivalent distilled water.The uteri samples were acquired on the day 8 of pregnancy.The numbers of implanted blastocysts on each side were counted.The endometrial protein expression of progesterone receptor(PR),Stat3 and p-Stat3 was deteected by Western Blot.The PR mRNA expression was detected by reverse transcription-polymerase chain reaction(RTPCR).Results:In the SI and SI+BR group,when compared with the control side,the number of implanted blastocyst in the side of Stat3 inhibitor reduced significantly(P<0.01),protein expression of PR and p-Stat3/Stat3 decreased significantly and PR mRNA expression decreased significantly(P<0.01).There was no significant difference in the number of implantation numbers,the expression of PR,p-Stat3/Stat3 protein and PR mRNA between the two groups in the control side and the Stat3 inhibitor side.Conclusion:The mechanism of Bushen Yiqi Huoxue Formula in improving embryonic implantation in rats may be related to its activation of Stat3 and up-regulation of PR expression and improvement of endometrial receptivity.
