摘 要:目的:探讨补阳还五汤对脊髓缺血再灌注损伤(SCII)后细胞自噬相关蛋白Beclin 1、微管相关蛋白1轻3 (LC3)、P62影响的机制。方法:48只SD大鼠随机分为假手术组、模型组、观察组和3-甲基腺嘌呤(3-MA)组。模型组腹腔内注射生理盐水,观察组在模型制备的前3天灌胃补阳还五汤,3-MA组腹腔注射3-MA注射液,然后夹闭腹主动脉40 min后开放。模型制备后,在缺血3 h、6 h后进行行为学评分、取材,免疫组化检测Beclin 1、LC3、P62的表达。结果:在缺血3 h后、6 h后,与假手术组比较,模型组行为学评分下降(P<0.05),Beclin 1、LC3、P62蛋白表达升高(P<0.05)。与模型组比较,观察组、3-MA组行为学评分升高,Beclin 1、LC3、P62蛋白表达降低(P<0.05)。在缺血3 h后,与3-MA组比较,观察组行为学评分及P62蛋白表达较高(P<0.05);在缺血6 h后,与3-MA组比较,观察组行为学评分较高(P<0.05),Beclin 1、LC3、P62蛋白表达较低(P<0.05)。结论:补阳还五汤通过调节Beclin 1、LC3和P62蛋白的表达,不仅可以促使细胞自噬发生,加快消化、降解受损、衰老、失能的细胞,给细胞的再生、重建提供原料,而且可以抑制细胞自噬过度发生,减少细胞凋亡程序,从而达到双向调节,保护神经细胞的目的。Objective: To discuss the mechanism of the effect of Buyang Huanwu tang on autophagy-related proteins including Beclin 1,microtubule-associated protein 1 light chain 3(LC3) and P62 after spinal cord ischemia-reperfusion injury(SCII). Methods: A total of 48 SD rats were randomly divided into the sham operation group, the model group, the observation group and the 3-Methyladenine(3-MA) group. Rats in the model group were given intraperitoneal injection of saline;rats in the observation group were given Buyang Huanwu tang by gavage three days before model preparation;rats in the 3-MA group were given intraperitoneal injection of 3-MA,and then the abdominal aorta was clamped for 40 minutes.After model preparation,the rats were undergoing behavior scoring and materials drewing after ischemia for three and six hours respectively,and the expression of Beclin 1,LC3 and P62 was detected by immunohistochemistry. Results:After ischemia for three and six hours respectively,compared with those in the sham operation group,the behavioral score in the model group was decreased(P<0.05), and the expression of Beclin 1, LC3 and P62 proteins was increased(P<0.05).Compared with those in the model group, the behavioral scores in the observation group and the 3-MA group were increased,and the expression of Beclin 1,LC3 and P62 proteins was decreased(P<0.05). After ischemia for three hours,the behavioral score and expression of P62 protein in the observation group were higher than those in the 3-MA group(P<0.05). After ischemia for six hours,the behavioral score in the observation group was higher than that in the 3-MA group(P<0.05), and the expression of Beclin 1, LC3 and P62 proteins was lower(P<0.05). Conclusion: By regulating the expression of Beclin 1, LC3 and P62 proteins, Buyang Huanwu tang can not only promote the occurrence of autophagy,accelerate digestion,degrade the damaged,aging and disabled cells and provide raw materials for cell regeneration and reconstruction,but also inhibit the excessive occurrence of autophagy and re
