摘  要:目的:探讨藏药镰形棘豆总黄酮对佐剂性大鼠关节炎白细胞介素1β(interleukin-1β,IL-1β)、肿瘤坏死因子α(tumor necrosis factorα,TNF-α)、环氧合酶1(cyclooxygenase-1,COX-1)及环氧合酶2(cyclooxygenase-2,COX-2)的影响。方法:将50只成年Wistar大鼠(雌雄各半)按性别匹配分为正常对照组、模型组、甲氨蝶呤组及镰形棘豆总黄酮低、高剂量组,以完全弗氏佐剂型(AA)关节炎大鼠为模型组。采用ELISA法检测大鼠血清IL-1β、TNF-α、COX-1及COX-2抗体水平,观察各组大鼠一般状态、AI积分及足关节肿胀情况。结果:与模型组及正常对照组相比,甲氨蝶呤及镰形棘豆总黄酮低、高剂量均能改善大鼠一般症状及足部肿胀程度,并能降低血清IL-1β、TNF-α及COX-2水平,差异具有统计学意义(P<0.05),但3组间比较差异无统计学意义(P>0.05);COX-1无明显变化(P>0.05)。结论:藏药镰形棘豆总黄酮能降低佐剂性关节炎大鼠血清IL-1β、TNF-α及COX-2水平,减轻病变关节损伤积分,有望作为治疗类风湿性关节炎的有效药物应用于临床。Objective: To explore the effects of total flavonoids of Lianxing Jidou(Oxytropis falcata Bunge) on IL-1β, TNF-α, COX-1 and COX-2 in adjuvant-induced arthritis rats. Methods: Fifty adult Wistar rats(half male and female) were allocated to normal control group, the model group, methotrexate group, low and high doses groups of Lianxing Jidou according to gender matching, rats were prepared into adjuvant-induced arthritis(AA),ELISA method was adopted to detect the levels of IL-1β, TNF-α, COX-1 and COX-2 in the rats, to obsevoe general situations, AI integrals, and foot swelling degrees of the rats in different groups. Results: Compared with the model group and normal control group, methotrexate, low and high doses of total flavonoids of Lianxing Jidou could improve common symptoms, foot swelling degrees, and lower the levels of IL-1 β, TNF-α and COX-2, and the difference showed statistical meaning(P<0.05), while the difference had no statistical meaning among three groups(P>0.05);no obvious change of COX-1 has been found(P>0.05). Conclusion: Total flavonoids of Lianxing Jidou could lower the levels of IL-1β, TNF-α and COX-2 in AA rats, relieve joint lesion integrals, and it is expected to be an effective drug in the treatment of rheumatoid arthritis.
