摘 要:目的建立白蔹药材中大黄素含量的测定方法,为完善白蔹药材的质量控制标准提供方法和依据。方法采用反相高效液相色谱法,色谱柱:Apollo-C18柱(4.6 mm×250 mm,5μm);流动相:甲醇-0.2%磷酸(85:15);流速:1.0 ml/min;柱温:室温;检测波长:220 nm。结果在选定的色谱条件下,白蔹药材中大黄素在0.124~3.968μg/ml范围内线性关系良好,直线回归方程为Y=53962X–966.46,r=0.9997,平均回收率为99.7%,RSD为2.5%(n=9)。结论该方法所测结果准确、灵敏度高、重现性好,适用于白蔹药材中大黄素的含量测定。Objective To establish a method for Emodin assay in Ampelopsis radix,and to improve the quality control of Ampelopsis radix.Methods Reversed phase high performance liquid chromatography was used.The column:Apollo-C18(4.6 mm×250 mm,5μm);mobile phase:methanol:0.2%phosphoric acid(85:15);flow rate:1.0 ml/min;column temperature:room temperature;detection wavelength:220 nm.Results With the specified chromatographic conditions,there was a good linear relationship for Emodin in Ampelopsis radix in the range of 0.124-3.968μg/ml.The linear regression equation was Y=53962X–966.46,r=0.9997.The average recovery was 99.7%.RSD=2.5%(n=9).Conclusion This method is accurate,sensitive and repeatable.It is suitable for the determination of emodin in Ampelopsis radix.
