摘 要:目的研究传统民族医学文献中用于治疗白癜风的诃子、牛柑子(即余甘子)、柴胡及药群组合(含木香、胡椒、干姜等)的药物血清对小鼠B16黑素瘤细胞黑素合成及酪氨酸酶活性的影响。方法采用"通法"制备上述药物高、低不同剂量的大鼠含药血清及不含药物的对照血清,以体外培养的小鼠B16黑素瘤细胞为模型,采用MTT法测定受试血清对B16B16黑素瘤细胞增殖活性的影响,NaOH细胞裂解法测定黑素合成量,L-DOPA氧化法测定对酪氨酸酶活性的影响。结果与正常对照组(含10%小牛血清)相比,无药血清对照组对B16黑素瘤细胞的增殖活性无影响(P>0.05);与无药血清对照组相比,除药群组合低剂量组外,其他受试血清均对小鼠B16黑素瘤细胞增殖活性无影响(P均>0.05)。与无药血清对照组相比,柴胡和药群组的高、低剂量组含药血清均对B16黑素瘤细胞黑素合成有促进作用,诃子低剂量、牛柑子高剂量组含药血清具有促进黑素合成的作用(P均<0.05)。与无药血清对照组相比,四种受试药物的高低剂量组含药血清对小鼠B16黑素瘤细胞酪氨酸酶活性均有激活作用(P均<0.05)。结论四种含药血清除组合低剂量组外,均可不同程度地促进黑素合成,此作用与提高酪氨酸酶活性有一定关系。Objective The study is to explore the effects of 4 kinds of serums containing Fructus chebulae,Phyllanthus emblica L.,Radix bupleuri,or composition(including radix aucklandiae,pepper,and ginger)on melanin synthesis and tyrosinase activity in murine B16 melanorma cells.The herbs in this study have been used clinically to leukodermia in medicine for years.Methods B16 melanorma cells were cultured being treated with herbs serums in high and low doses respectively in vitro.MTT was tested to evaluate effects of the herbs on the proliferation of cells,Na OH cell lysis was used to test the melanin synthesis,L-DOPA oxidation was tested to check the tyrosinase activity effects.Results Compared with the normal control group,pharmacal serums were no significantly difference on the effects of proliferation of B16 melanorma cells(P>0.05),except the composition in low dose group.Radix bupleuri in high and low doses,pharmaceutical composition in high and low doses,fructus chebulae in low dose(P<0.05),and Phyllanthus emblica L.in high dose were significantly increased the levels of melanin synthesis in B16 melanorma cells when compared with the blank control group respectively(P all<0.05).All the treatment groups were significantly increased the levels of tyrosinase activity when compared with the blank control group(P all<0.05).Conclusion The 4 herb-serums can promote the levels of melanin synthesis in B16 melanorma cells with no effects on proliferation,which is related to the improvement of tyrosinase activity.
