摘  要:目的 在四逆汤的基础上建立附子炮制新方法.方法 附子饮片25%氨水提取物的分析采用Phenomenex RPC18色谱柱(250 mm×4.6 mm,5μm);流动相0.1%二乙胺-乙腈,梯度洗脱;柱温25℃;体积流量1.0 mL/min;检测波长230 nm.分别考察炮制过程中所用不同生物酶比例及用量、甘草与干姜的用量以及蒸制时间对附片生物碱含有量的影响,进而筛选出最优的炮制工艺.结果 6种生物碱在各自范围内线性关系良好(r>0.997 5),平均加样回收率91.51%~ 102.3%,RSD 0.90%~2.41%.在炮制过程中,加入混合酶,随后使用甘草粉-干姜粉的混合粉均匀挂粉后进行高压蒸制40 min,得到效毒比最高的新型附片.此新型附片的苯甲酰新乌头原碱含有量可达0.035%,而双酯型生物碱总量低至0.010%.结论 该方法简便易行可控,所得附片有效且可安全使用,可作为替代附子传统加工工艺的新方法.AIM To establish a new method of processing Aconiti Lateralis Radix Praeparata based on Sini De coction.METHODS The analysis of 25%ammonia extract from Aconiti Lateralis Radix Praeparata was developed on a 25℃thermostatic Phenomenex RP C18column(250 mm×4.6 mm,5μm),with the mobile phase comprising of 0.1%diethylamine acetonitrile flowing at 1 mL/min in a gradient elution manner,and the detection wavelength was set at 230 nm.An optimal process was screened out.The parameters included the proportion and dosage of enzymes,the dosage of liquorice powder and ginger powder,and the steaming time.RESULTS Six al kaloids showed good linear relationships within their own ranges(r>0.9975),whose average recoveries were 91.51%-102.3%,with the RSDs of 0.90%-2.41%.After treating with mixed enzyme powder,Aconiti Lateralis Radix Praeparata was mixed with glycyrrhiza and ginger powder,then steamed at high pressure for 40 minutes.In the new Aconiti Lateralis Radix Praeparata processed pieces,the content of benzoylmes aconine was 0.035%,and the total amount of diester alkaloids was as low as 0.010%.CONCLUSION The simple and available method can be used as a new method for replacing the traditional processing technology of Aconiti Lateralis Radix Praeparata.
