摘  要:目的分析葡萄糖激酶调节蛋白基因rs780094多态性与冠心病痰瘀互阻证之间的关系。方法收集206例冠心病病人病史及实验室数据资料,提取DNA测序并进行rs780094多态性分析。依据是否为痰瘀互阻证将病人分为两组,进行统计分析。结果痰瘀互阻证冠心病病人较非痰瘀互阻证病人的血糖稍高[(6.09±2.36)mmol/L与(5.98±2.13)mmol/L,P=0.574],但未达到统计学意义。冠心病痰瘀互阻组中,rs780094 CC基因型的比例高于非痰瘀互阻证组(28.75%与14.29%,P=0.013),其等位基因C的比例也高于非痰瘀互阻证组(53.75%与39.68%,P=0.006)。Logistic回归结果显示,rs780094 CC基因型与冠心病痰瘀互阻证显著相关。结论冠心病痰瘀互阻证型与rs780094多态性存在显著关联,其分子机制有待进一步研究。Objective To analyze the relationship between rs780094 polymorphism of glucokinase regulatory protein gene and phlegm-blood stasis syndrome in coronary heart disease(CHD).Methods General population information and data from clinical lab of 206 patients with CHD were collected.Rs780094 polymorphism were analyzed by DNA sequencing of the whole blood samples.According to whether the phlegm-blood stasis syndrome existed,the patients were divided into two groups,and statistical analysis was carried out.Results The fasting blood glucose concentration in patients with phlegm-blood stasis syndrome was slightly higher than that in patients without phlegm-blood stasis syndrome[(6.09±2.36)mmol/L vs(5.98±2.13)mmol/L,P=0.574],but the difference was not significant.The proportion of rs780094 CC genotype in CHD patients with phlegm-blood stasis syndrome was significantly higher than that in CHD patients with non-phlegm-blood stasis syndrome(28.75%vs 14.29%,P=0.013).The proportion of allele C was also significantly higher than that in non-phlegm-stasis syndrome group(53.75%vs 39.68%,P=0.006).Logistic regression results showed that rs780094 CC genotype was significantly correlated with the phlegm-blood stasis syndrome of coronary heart disease.Conclusion The Phlegm-blood stasis syndrome in CHD is significantly associated with rs780094 polymorphism,and its molecular mechanism needs further study.
