摘 要:本文旨在检测不同浓度氯化钠胁迫对高效降酚菌热带假丝酵母菌株GY8降解苯酚性能的影响。通过在含有0.1,10,25,50,100 g·L^-1氯化钠的无机盐培养基中培养GY8菌,检测不同氯化钠浓度胁迫对菌OD600值、降解苯酚速率,降酚关键酶的活性的影响。结果表明,GY8菌在含0~100 g·L^-1盐浓度培养基中均可生长,且都有降酚活性。但随着盐浓度增加,GY8菌OD600值和降酚速率及邻苯二酚1,2-双加氧酶酶比活力均呈现抑制趋势,苯酚羟化酶的活性先增强后抑制。当氯化钠浓度超过25 g·L^-1时,抑制作用明显增强。通过本研究可以得出GY8菌可耐受0~100 g·L^-1氯化钠,且氯化钠对GY8降酚活性具有梯度抑制作用。The aim of this study was to investigate the influence of different concentration of sodium chloride stress on phenol-degrading activity of Candida tropicalis GY8.GY8 strain were cultured in the inorganic salt medium containing 0.1,10,25,50 and 100 g·L^-1 sodium chloride.The effects of different concentrations of sodium chloride on the OD600 value,the phenol-degrading rate,and the key enzyme activities for phenol degradation were detected.The results showed that GY8 strain could grow in the culture medium containing 0~100 g·L^-1 salt concentration,and all of them had the activity of phenol degradation.However,with the increase of salt concentration,the OD600 value,the phenol-degrading rate and the specific activity catechol 1,2-dioxygenase of GY8 strain showed inhibitory trend.The activity of phenol hydroxylase increased first and then inhibited.When the concentration of sodium chloride was more than 25 g·L^-1,the inhibition was obviously enhanced.This study concluded that GY8 strain can tolerate 0~100 g·L^-1 salt concentration,and NaCl has a gradient inhibitory effect on the phenol-degrading activity of GY8.
