摘  要:目的探讨儿童支原体肺炎感染的Th17/调节性T细胞(Treg)细胞水平变化及白细胞介素-6(IL-6)/信号转导和转录激活子3(STAT3)、白细胞介素-2(IL-2)/信号转导和转录激活子5(STAT5)信号通路作用。方法选择2017年6月-2018年12月海南省妇女儿童医学中心医院收治的儿童支原体肺炎感染患儿107例作为研究对象,其中急性期49例,恢复期58例。另选择2017年6月-2018年12月健康体检小儿50名作为对照组。采用流式细胞术检测Th17和Treg细胞水平,采用酶联免疫吸附法测定血清IL-2和IL-6水平,采用实时荧光定量PCR术检测STAT3mRNA和STAT5mRNA表达。结果急性期组Th17和Th17/Treg高于恢复期组和对照组,而Treg%低于恢复期组和对照组(P<0.05);恢复期组Th17和Th17/Treg高于对照组,而Treg低于对照组(P<0.05)。急性期组血清IL-2水平低于恢复期组和对照组,而血清IL-6水平高于恢复期组和对照组(P<0.05);恢复期组血清IL-2水平低于对照组,而血清IL-6水平高于对照组(P<0.05)。急性期组STAT3mRNA高于恢复期组和对照组,而STAT5mRNA低于恢复期组和对照组(P<0.05);恢复期组STAT3mRNA高于对照组,而STAT5mRNA低于对照组(P<0.05)。结论Th17/Treg及IL-6/STAT3、IL-2/STAT5信号通路参与了儿童支原体肺炎感染发生、发展,支原体肺炎感染患儿体内存在Th17与Treg细胞亚群失衡,存在IL-6/STT3信号通路激活,IL-2/STAT5信号通路活性下调。OBJECTIVE To investigate the variation level of Th17/regulatory T cells(Treg)in children with Mycoplasma pneumonia and the function of interleukin-6(IL-6)/signal transduction and activator of transcription 3(STAT3),interleukin-2(IL-2)/signal transduction and activator of transcription 5(STAT5)signaling pathway.METHODS The total of 107 children with M.pneumonia infection admitted to our hospital from June 2017 to December 2018 were recruited as the study subjects,including 49 in acute stage and 58 in convalescent stage.Another 50 healthy children during physical examination from June 2017 to December 2018 were enrolled as the control group.The expression levels of Th17 and Treg in cells were detected by flow cytometry assay,serum IL-2 and IL-6 were measured by ELISA,and the expression of STAT3 and STAT5 mRNA was detected by real-time fluorescence quantitative PCR.RESULTS Th17 and Th17/Treg in acute phase group were significantly higher than that in the convalescence group and the control group,whereas Treg in the in acute phase group was significantly lower than that in the convalescence group and control group(P<0.05).Th17 and Th17/Treg in the convalescence group were significantly higher than that in the control group,whereas Treg was significantly lower than that in the control group(P<0.05).Serum IL-2 level in the acute phase group was lower than that in the convalescence group and control group,and serum IL-6 level was higher than that in the convalescence group and control group(P<0.05);serum IL-2 level in the convalescence group was significantly lower than that in the control group,and serum IL-6 level was significantly higher than that in the control group(P<0.05).The expression of STAT3 in the acute phase group was significantly higher than that in the convalescent phase group and control group,and the expression of STAT5 mRNA was significantly lower than that in the convalescent phase group and control group(P<0.05);the expression of STAT3 in the convalescent phase group was higher than that in the cont
