摘 要:目的:探讨准噶尔乌头生物碱及炮制水解产物抗肿瘤活性研究,并对准噶尔乌头生物碱及其水解产物的细胞毒活性进行初步研究。方法:采用CCK-8法检测不同浓度准噶尔乌头生物碱及炮制水解产物作用不同时间后A549人肺癌细胞的增殖情况,筛选出抗肿瘤有效的准噶尔乌头单体。结果:准噶尔乌头生物碱及炮制水解产物作用于A549人肺癌细胞呈现出增殖抑制作用,且呈浓度依赖性(P<0.01),准噶尔乌头碱对A549人肺癌细胞有抑制作用,干预24,48,72 h时半抑制浓度(IC50)分别为636.791,597.083,544.382μg·ml^-1。结论:研究表明准噶尔乌头生物碱及炮制水解产物可抑制A549人肺癌细胞生长。Objective:To study the antitumor activity of aconitum alkaloids and processed hydrolysates in Junggar,and study the cytotoxicity of aconitine and its hydrolysates as well.Methods:CCK-8 assay was used to detect the proliferation of A549 human lung cancer cells treated with different concentrations of aconitine alkaloids and processed hydrolysates in Junggar for different duration,and the effective anti-tumor aconitine monomer was screened.Results:Aconitine alkaloids and processed hydrolysates in Junggar showed inhibitory effect on the proliferation of human lung cancer cells.Aconitine in Junggar could inhibit the proliferation of human lung cancer cells,and it was concentration-dependent(P<0.01).IC50 was 636.791,597.083 and 544.382μg·ml^-1 after the intervention for 24,48 h and 72 h,respectively.Conclusion:Aconitine alkaloids and processed hydrolysates in Junggar can inhibit the growth of human lung cancer cells.
