摘  要:通过对10份枸杞属种质材料的叶绿体基因trnL-trnF进行克隆测序,获得10条DNA序列,经过比对分析及构建系统发育树,得出以下结果:①trnL-trnF序列总长为1156 bp,其中保守位点1146个,变异位点10个,包括简约信息位点9个,自裔位点1个;GC含量均值为35.9%;碱基转换颠换值为4.4。②获得的系统发育树分成了两大支,即黑果、W-12-30、黄果变和W-12-27聚为一大支,其余品种聚为一支,各分支内部均具有较高的自展支持率。因此,trnL-trnF序列可以作为初步鉴定枸杞属植物的DNA条形码。Through cloning for sequencing the chloroplast genes trnL-trnF from 10 germplasm materials of Lycium L.,10 DNA sequences were obtained.After alignment analysis and building phylogenetic tree,the results showed that①the total length of trnL-trnF sequence was 1156 bp,which included 1146 conserved sites,10 variable sites,9 PI-parsimony-informativesites and a C-conservedsite.The mean value of GC content was 35.9%and the R value was 4.4.②The MP tree divided 10 test materials into two parts.Heiguo,W-12-30,Huangguobian and W-12-27 plants were divided into one branch.Others were in one branch.High favorability rating of bootstrap was showed in two parts.Therefore,trnL-trnF sequence could be the DNA barcode to preliminarily distinct Lycium L.plants.
