摘 要:目的:观察小柴胡汤对肝郁脾虚型功能性消化不良模型大鼠胃、脊髓、下丘脑组织中胆囊收缩素(CCK)和降钙素基因相关肽(CGRP)蛋白水平的影响,探讨小柴胡汤治疗肝郁脾虚型功能性消化不良的可能机制。方法:将48只SD大鼠随机分为空白组、模型组、小柴胡汤组、多潘立酮组,每组12只,雌雄各半。后3组采用复合病因造模法制作肝郁脾虚型功能性消化不良模型。依据大鼠体质量计算灌胃药量,小柴胡汤组大鼠药量为每日7.29 g/kg,多潘立酮组为每日2.7 mg/kg,空白组和模型组灌服同等体积的生理盐水,治疗14 d后取材,采用Western Blotting方法检测各组大鼠胃、脊髓、下丘脑组织中CCK、CGRP蛋白水平。结果:与模型组和多潘立酮组相比,小柴胡汤组大鼠胃CCK蛋白水平、胃和脊髓组织中CGRP蛋白水平均明显提高,差异均有统计学意义(P<0.05)。结论:小柴胡汤可改善大鼠胃、脊髓组织中CCK和CGRP的表达,这可能是小柴胡汤治疗肝郁脾虚型功能性消化不良的作用机制之一。Objective:To observe the effect of Xiaochaihu Decoction(小柴胡汤)on the protein levels of cholecystokinin(CCK)and calcitonin gene-related peptide(CGRP)in the stomach,spinal cord and hypothalamus of rats with functional dyspepsia of liver depression and spleen deficiency pattern and to explore the possible mechanism of Xiaochaihu Decoction in the treatment of functional dyspepsia of liver depression and spleen deficiency pattern.Methods:Total 48 SD rats were randomly divided into four groups:blank group,model group,Xiaochaihu Decoction group and domperidone group,with 12 rats in each group,half male and female.The functional dyspepsia of liver depression and spleen deficiency pattern was made by compound pathogeny modeling in the latter 3 groups.The amount of gavage was calculated ac cording to the body weight of the rats.The Xiaochaihu Decoction group had a daily dose of 7.29 g/kg and the domperidone group had a daily dose of 2.7 mg/kg.After 14 days,samples were taken and the levels of CCK and CGRP in the stomach,spinal cord,and hypothalamus of model rats were detected by Western Blotting methods.Results:Compared with the model group and domperidone group,the level of gastric CCK protein and the level of CGRP protein in the stomach and spinal cord tissue of rats in the Xiaochaihu Decoction group were significantly increased,the difference was statistically significant(P<0.05).Conclusions:Xiaochaihu Decoction can improve the expression of CCK and CGRP in the stomach and spinal cord tissues of rats,suggesting that this may be One of the mechanisms of Xiaochaihu Decoction in treating functional dyspepsia of liver depression and spleen deficiency pattern.
