摘 要:目的探讨间歇低氧对大鼠海马神经细胞突触相关蛋白表达及认知功能的影响。方法选择成年Wistar雄性大鼠64只,采用随机数字法分为对照组32只和5%间歇低氧组32只,每组又分为7、14、21、28 d 4个时间点,每个时间点8只。采用Morris水迷宫对2组大鼠行学习记忆功能检测;透射电镜观察大鼠海马CA1区神经细胞突触数量及相关结构变化;免疫印迹法检测海马CA1区神经细胞突触素蛋白(SYP)和突触后致密蛋白95(PSD95)表达。结果与对照组比较,5%间歇低氧组7、14、21、28 d逃避潜伏期明显延长,穿越目标象限时间明显缩短(P<0.05)。5%间歇低氧组随时间增加逃避潜伏期明显延长,穿越目标象限时间明显缩短(P<0.05)。透射电镜下观察显示,与对照组比较,5%间歇低氧组7、14、21、28 d大鼠海马CA1区神经细胞突触数量减少,突触间隙不清,随间歇低氧时间的延长,突触损伤越重;与对照组比较,5%间歇低氧组7、14、21、28 d海马神经细胞SYP和PSD95表达明显减少(0.60±0.04 vs 0.78±0.02,0.49±0.03 vs 0.76±0.03,0.27±0.02 vs 0.78±0.03,0.19±0.02 vs 0.77±0.02,P<0.05;0.39±0.02 vs 0.50±0.07,0.28±0.03 vs 0.52±0.02,0.21±0.04 vs 0.53±0.06,0.11±0.02 vs 0.50±0.08,P<0.05)。5%间歇低氧组随时间增加SYP和PSD95表达明显减少(P<0.05)。结论间歇低氧可引起认知功能减退,可能与其抑制大鼠海马神经细胞突触相关蛋白SYP和PSD95表达相关。Objective To study the effect of intermittent hypoxia on expression of hippocampal synapse-related proteins and cognitive function in rats.Methods Sixty-four adult Wistar male rats were randomly divided into control group(n=32) and 5% intermittent hypoxia group(n=32).The animals in each group were further divided into 7 d group,14 d group,21 d group and 28 d group(8 in each group).The learning and memory functions in two groups were tested by Morris water maze test.The number of synapses and change of related structures in hippocampal CA1 area were detected by transmission electron microscopy.The expressions of synaptophysin(SYP) and PSD95 in hippocampal CA1 neurons were detected by Western blot.Results The escape latency was significantly longer while the target quadrant crossing time was significantly shorter in 5% intermittent hypoxia group on days 7,14,21 and 28 than in control group(P<0.05).The escape latency time increased significantly while the target quadrant crossing time decreased significantly with the increasing time in 5% intermittent hypoxia group(P<0.05).Transmission electron microscopy showed that the number of neuron synapses in hippocampal CA1 area was significantly smaller,the synaptic gap was significantly unclear,and the damage of synapses was significantly severer in 5% intermittent hypoxia group on days 7,14,21 and 28 than in control group(P<0.05).The expression levels of SYP and PSD95 were significantly lower in 5% intermittent hypoxia group on days 7,14,21 and 28 than in control group(0.60±0.04 vs 0.78±0.02,0.49±0.03 vs 0.76±0.03,0.27±0.02 vs 0.78±0.03,0.19±0.02 vs 0.77±0.02,P<0.05;0.39±0.02 vs 0.50±0.07,0.28±0.03 vs 0.52±0.02,0.21±0.04 vs 0.53±0.06,0.11±0.02 vs 0.50±0.08,P<0.05).The expression levels of SYP and PSD95 decreased significantly in 5% intermittent hypoxia group with the increasing time(P<0.05).Conclusion Intermittent hypoxia can decrease the cognitive function of rats by inhibiting the expressions of SYP and PSD95 in their hippocampal neurons.
