摘  要:目的:研制一种模块化三维灌注培养系统,解决目前培养系统结构复杂、难消毒、易污染的问题。方法:该系统主要由生物反应器模块、氧合模块、动力模块以及换液模块构成,各模块壳体通过3D打印制作。其中生物反应器模块主体为80 ml蓝口瓶,装载培养基及细胞支架复合物。氧合模块装载膜肺、空气过滤器、气泡过滤器等,为培养系统的核心组件。动力模块由蠕动泵和空气泵组成,为培养基及气体提供动力。换液模块中装有深紫外LED灯,可进行消毒和更换培养基。结果:通过该系统进行大鼠脾脏脱细胞支架培养,细胞与支架黏附紧密、生长状态良好,且未发生污染。结论:该系统具有结构合理、易消毒、污染少、制作成本低、稳定性高、易于扩展且操作简单的优点,可很方便地进行基于脱细胞支架的三维灌注培养,值得推广应用。Objective To develop a modular three-dimensional perfusion culture system to eliminate the shortcomings of the existing system in complicated structure,difficult disinfection,vulnerability to pollution and etc.Methods The system was composed of several modules for bioreactor,oxygenation,power and liquid changing,and all the shells were made by 3D printing.The bioreactor module had an 80 ml blue bottle as its main body,which was loaded with culture medium and cell scaffold complex.The oxygenation module was equipped with membrane lung,gas filter,bubble filter,etc.,which was the core component of the culture system.The power module consisted of a peristaltic pump and an air pump to provide power for the medium and gas.The liquid changing module included a deep UV LED lamp,which could be used to disinfect and change the medium.Results The system was used to culture the acellular scaffold of rat spleen,and the cells adhered to the scaffold tightly and grew well without pollution.Conclusion The perfusion culture system developed has the advantages of reasonable structure,easy disinfection,few pollution,low production cost,high stability,easy expansion and simple operation,and thus is worthy promoting for three-dimensional perfusion culture of decellularization scaffold.
