摘 要:目的探究并分析大肠杆菌K5多糖对糖尿病性视网膜病变(DR)小鼠视网膜损伤的保护作用及Bcl-2相关X蛋白(Bax)、核因子E2相关因子2(Nrf2)表达的影响。方法选取健康雄性C57BL/6小鼠120只,按照随机数字表法将其分为对照组和观察组,每组60只。两组小鼠进行糖尿病性视网膜病变造模处理,对照组小鼠又随机分为未处理组、处理(生理盐水)后1周组、处理(生理盐水)后2周组,每组20只。观察组小鼠又随机分为未处理组、处理(大肠杆菌K5多糖)后1周组、处理(大肠杆菌K5多糖)后2周组,每组20只。观察两组小鼠处理前、处理后1周、处理后2周视觉电生理指标及Bax、Nrf2蛋白表达水平。结果两组小鼠处理前波峰潜时、波峰振幅比较无显著差异(P>0.05),处理后,两组小鼠视觉电生理指标显著改善(P<0.05),观察组小鼠波峰潜时显著低于对照组(P<0.05),波峰振幅显著高于对照组(P<0.05);两组小鼠处理前视网膜节细胞、视网膜内网层细胞Bax蛋白水平比较无显著差异(P>0.05),处理后,两组小鼠视网膜节细胞、视网膜内网层细胞Bax蛋白水平显著降低,观察组视网膜节细胞、视网膜内网层细胞Bax蛋白水平明显低于对照组(P<0.05);两组小鼠处理前Nrf2蛋白水平比较无显著差异(P>0.05),处理后,两组小鼠Nrf2蛋白水平显著升高(P<0.05),观察组小鼠Nrf2蛋白水平明显高于对照组,差异具有统计学意义(P<0.05)。结论大肠杆菌K5多糖对DR小鼠视网膜损伤具有保护作用,同时能有效改善小鼠凋亡基因Bax及Nrf2的表达。Objective To investigate and analyze the protective effect of E.coli K5 polysaccharide on diabetic retinopathy(DR)mice retina injury and the expression of Bax and Nrf2.Methods 120 healthy male C57BL/6 mice were randomly divided into two groups:control group and observation group,60 mice in each group.The two groups were treated with diabetic retinopathy.The control group was randomly divided into untreated group,one week group after treatment(normal saline)and two weeks group after treatment(normal saline),20 mice in each group.The mice in the observation group were randomly divided into three groups:untreated group,treated group(E.coli K5 polysaccharide)for one week and treated group(E.coli K5 polysaccharide)for two weeks.The visual electrophysiological indexes,Bax and Nrf2 levels were observed before,1 week and 2 weeks after treatment.Results The visual electrophysiological indexes of the two groups were significantly different before and after treatment(P<0.05).There was no significant difference between the two groups before treatment(P>0.05).After treatment,the visual electrophysiological indexes of the two groups were significantly improved(P<0.05).The wave peak latency of the observation group was significantly lower than that of the control group(P<0.05),and the wave peak amplitude was significantly higher than that of the control group(P<0.05).There was no significant difference of Bax level between the two groups before and after treatment(P>0.05).After treatment,the Bax level of the retinal ganglion cells and the retinal inner reticulum cells in the two groups decreased significantly,and the Bax level of the retinal ganglion cells and the retinal inner reticulum cells in the observation group was significantly lower than that in the control group(P<0.05).There was no significant difference of Nrf2 level between the two groups before and after treatment(P>0.05).After treatment,the level of Nrf2 in the two groups decreased significantly(P<0.05).The level of Nrf2 in the observation group was significantly
