摘  要:[目的]研究藏药西伯利亚蓼不同部位降尿酸活性,并采用HPLC法进行分析检测。[方法]以氧嗪酸钾盐诱导建立小鼠急性高尿酸血症模型,连续灌胃给予西伯利亚蓼水部位(2.0、4.0 g/kg)、30%乙醇部位(2.0、4.0 g/kg)、50%乙醇部位(2.0、4.0 g/kg)、90%乙醇部位(2.0、4.0 g/kg)及别嘌呤醇(AP,0.01 g/kg)7 d,探讨西伯利亚蓼不同部位对急性高尿酸血症血尿酸(UA)、肝脏黄嘌呤氧化酶(XOD)活性及尿酸转运体(mURAT1、mGLUT9和mOAT1)mRNA表达的影响,并对不同部位进行HPLC分析。[结果]与模型组相比,AP以及30%、50%、90%乙醇部位显著降低高尿酸血症的血尿清水平;50%、90%乙醇部位既可以抑制黄嘌呤氧化酶活性又可以调控尿酸转运体的表达;30%乙醇部位仅对尿酸转运体具有调控作用;不同部位HPLC图显示除了30%、50%乙醇部位化学成分有重叠,西伯利亚蓼不同部位化学成分差别较大。[结论]30%、50%、90%乙醇部位是西伯利亚蓼的降尿酸的活性部位,其作用机制为抑制XOD活性和调控尿酸转运体的表达;西伯利亚蓼不同部位HPLC图有一定的差别,为探讨西伯利亚蓼不同部位降尿酸药效物质基础提供一定的依据。[Objective]The research aimed to study the lowering uric acid activity of different parts of Polygonum sibiricum,and to analyze and detect it by HPLC.[Method]Water fractions(2.0,4.0 g/kg),30%ethanol fractions(2.0,4.0 g/kg),50%ethanol fractions(2.0,4.0 g/kg),90%ethanol fractions(2.0,4.0 g/kg)and allopurinol(AP,0.01 g/kg)were administered to hyperuricemia mice induced by potassium oxonate for seven days.The level of uric acid(UA)and xanthine oxidase(XOD)on different fractions were analyzed and the mRNA express of renal protein transporter(mOAT1,mURAT1 and mGLUT9)were also studied.The different parts of P.sibiricum were analyzed by HPLC.[Result]Compared with the model group,AP and 30%,50%,90%ethanol fractions of P.sibiricum obviously lowered the serum UA levels;50%and 90%ethanol fractions not only inhibited the activity of XOD,but also regulated the expression of uric acid transporter;30%ethanol fractions only regulated the uric acid transporter.The HPLC chart of different parts showed that the chemical constituents of different fractions were obvious distinctions except for the overlap of 30%and 50%ethanol fractions.[Conclusion]30%,50%and 90%ethanol fractions are effective fractions of P.sibiricum and its mechanism is to inhibit XOD activity and regulate the expression of uric acid transporters.The HPLC images of different parts have certain differences,which provide a basis for exploring the physiochemical discussion for the reduction of uric acid.
