摘  要:目的:通过研究铝对大鼠学习记忆及e-fos基因表达的影响,探究铝损伤记忆功能的机制。方法:将Wistar大鼠按体重随机分为对照组(control)、低剂量染毒组(AI-L)、中剂量染毒组(AI-M)和高剂量染赤组(AI-H),染毒时限为大鼠出生当天(1 d)至90 d。染毒结束后,通过Morris水迷宫检测大鼠的学习记忆能力,石墨炉原子吸收光谱法检测大鼠海马铝含量,HE染色法观察海马CA3区神经细胞形态、RT-PCR检测大鼠海马c-fos基因表达,免疫组化法检测大鼠海马Fos蛋白表达。结果:随着铝暴露剂量增加,大鼠空间记忆能力降低,海马CA3区神经细胞形态受损加重,海马铝含量增加,海马CA3区c-fosmRNA表达降低,海马CA3区Fos蛋白含量降低。结论:亚慢性铝暴露损伤大鼠空间记忆功能,其机制可能与海马CA3区c-fos mRNA表达和蛋白含量降低有关。Objective:To explore the mechanisms of memory function impaired by Alumi-num,by investigating the effects of aluminum on learning memory and c-fos gene expression inrats.Methods:W istar rats were randomly divided into control group and low dose group(Al-L),middle dose group(Al-M)and high dose group(Al-M)according to body weight,r-ats were exposed from the partal day(1 d)to 90 d,lasting 90 days.Afer exposure,learning and memory ability of rats was dedetected by Morris water maze,hippocampal aluminum content was dected by graphite furmace atomie absorption spectrometry(GFAAS),neuron morphology in hippocampal CA3 area was observed by HE stain,hip-pocampal e-fos gene expression was detected by RT-PCR and hippocampal Fos protein content of rats was determined by immunohistochemistry.Results:Learning memory function and neurond cell morphology of rats was impaired,the aluminum content in hippocampus was increased,cfos mRNA expression and Fos protein content in hippocampal CA3 area were decreasedwith the increasing aluminum dose.Conclusion:Aluminum impairs learming and memory in rats,which may be related to the decrease of c-fos mRNA expression and Fos protein expression in hippocampal CA3 area.
