摘 要:目的探讨体外培养骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)对大鼠慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)治疗产生的影响。方法进行外体提取3周龄雄性SD大鼠的BMSCs原代细胞,培养到第3代用于实验。将60只SD雌性大鼠随机等分为3组:空白对照组、COPD模型组以及BMSCs干预治疗组。通过持续吸烟法建立COPD大鼠模型后,模型建立成功14天后,治疗组尾静脉给药方式给予BMSCs(1×10~6/ml)治疗,空白对照组和COPD模型组给予相同体积的生理盐水。通过HE染色法检验肺组织形态并验证模型是否成功,免疫组织化学和Western blotting检测TNF-α和IL-8蛋白的分布和表达。结果 HE结果显示,模型组与对照组相比,肺组织炎性细胞浸润、肺泡壁破坏更为显著,治疗组的情况得到了一定程度的缓解。免疫组化结果显示,对照组的TNF-a和IL-8蛋白仅有微量的基础表达,模型组的蛋白表达明显高于对照组,治疗组相比较模型组,蛋白表达出现下降趋势,但比对照组的基础表达仍有明显增高。Western blotting的结果显示,TNF-α和IL-8蛋白在对照组中只有少量的基础表达,模型组的蛋白出现了明显的上升,治疗组的蛋白出现下降,但是依然比对照组的表达高。结论在大鼠慢性阻塞性肺疾病的发病过程中,给予BMSCs干预治疗,能够有效抑制炎性因子的表达,进而缓解COPD病理进程。Objective To study the effect of exogenous BMSCs transplantation on COPD treatment in rats. Methods The BMSCs of male SD rats in 3 weeks old were isolated and cultured in vitro and transferred to the 3 generation. 60 SD female rats were randomly divided into 3 groups: the control group, the COPD model group and the BMSCs treatment group. After the COPD rat model was established by continuous fumigation, the model was successfully established for 14 days. The treatment group was treated with BMSCs(1 x10^6/ml), and the control group and the model group were given equal dose of NS. The morphological changes of lung tissue were observed by HE staining, and the distribution and expression of TNF-α and IL-8 proteins were detected by immunohistochemistry and Western blotting. Results The results of HE showed that the inflammatory cell infiltration and alveolar wall destruction in the lung tissue were more significant compared with the control group, and the treatment group was relieved to a certain extent. The results of immunohistochemistry showed that the protein expression of TNF-α and IL-8 in the control group was only a small amount of basic expression, and the protein expression in the model group was significantly higher than that in the control group. The protein expression in the treatment group was lower than that in the model group, but it was still higher than the basic expression of the control group. The results of Western blotting showed that only a small amount of basic expression of TNF-α and IL-8 protein in the control group, the expression of the model group increased significantly, and the expression of the treatment group decreased, but was higher than the control group. Conclusions In the process of chronic obstructive pulmonary disease in rats, the intervention of BMSCs can effectively inhibit the expression of inflammatory factors and then alleviate the pathological process of COPD.
