摘 要:【目的】从调控肠道微生物角度探讨通腑降浊方对便秘型肠易激综合征(IBS-C)大鼠的效应机制。【方法】将49只雄性SD大鼠随机分为7组,即正常组,模型组,西沙必利组,益生元组,中药高、中、低剂量组,每组7只。采用冰水灌胃法复制IBS-C大鼠模型。造模成功后,西沙必利组给予西沙必利片(剂量为3.6 mg·kg-1·d-1)灌胃,益生元组给予乳果糖口服溶液(剂量为1667.5 mg·kg-1·d-1)灌胃,中药高、中、低组分别给予通腑降浊方颗粒(剂量分别为18.500、9.250、4.625 g·kg-1·d-1)灌胃治疗,模型组和正常组给予等量生理盐水灌胃。治疗14 d后,留取大鼠粪便,通过16S rDNA扩增子测序技术,分析其菌群特征。采用苏木素-伊红(HE)染色法观察结肠组织病理变化。【结果】与正常组比较,模型组及各治疗组大鼠出现便秘、易激惹,粪便菌群结构紊乱、丰富度下降;与模型组比较,各治疗组大鼠粪便性状及情绪好转,粪便菌群结构接近正常组,其中中药高剂量组治疗后作用最明显。【结论】通腑降浊方可能通过提高IBS-C大鼠肠道微生物丰富度,调整菌群紊乱,稳定肠道微生态发挥对IBS-C大鼠的治疗作用。Objective To explore the effect of Bowels-Unblocking and Turbidity-Purging Recipe on microbial communities in faeces of rats with constipation-predominant irritable bowel syndrome(IBS-C). Methods Fortynine male SD rats were randomly divided into 7 groups,namely normal group,model group,cisapride group,probiotics group,and high-,middle-and low-dose Chinese medicine groups,7 rats in each group. The rat model of IBS-C was established by intragastric administration of ice water. After successful modeling,the western medicine group was given intragastric administration of cisapride tablets(at the dosage of 3.6 mg·kg-1·d-1),the prebiotics group was given intragastric administration of lactulose oral solution(at the dosage of 1 667.5 mg·kg-1·d-1),the high-, middle-and low-dose Chinese medicine groups were given intragastric administration of BowelsUnblocking and Turbidity-Purging Recipe Granules(at the dosage of 18.500, 9.250, 4.625 mg ·kg-1·d-1,respectively),and the model group and normal group were given intragastric administration of the same volume of normal saline. After 14 days of treatment, the microbial communities in faeces was detected by 16S rDNA augmented sequencing technology to analyze the characteristics. The histopathological feature of colon tissues was observed by hematoxylin-eosin(HE)staining method. Results Compared with the normal group,the symptoms of constipation and irritability,and the disorder and decreased richness of microbial communities in faeces were seen in the model group and various treatment groups. Compared with the model group,the fecal characteristics and mood of the rats in various treatment groups were improved,and the fecal microbial communities was close to the normal group,among which the effect was most obvious in the high-dose Chinese medicine group.
