摘  要:【目的】基于Janus激酶2(JAK2)/信号传导及转录激活因子3(STAT3)信号通路探讨大承气汤对重症急性胰腺炎(SAP)大鼠肠道的保护作用。【方法】采用3.5%牛磺胆酸钠逆行胆胰管内注射制备SAP大鼠模型。将造模成功的大鼠随机分为模型组、JAK2抑制剂组(术前2 h给予鲁索替尼灌胃)、STAT3抑制剂组(术前2 h给予Stattic腹腔注射)、大承气汤组(术前2 h给予大承气汤灌胃),另设假手术组(逆行胆胰管内注射生理盐水);再将各组大鼠分为造模后3、6、12、18 h亚组。采用苏木素-伊红染色观察末端回肠组织的病理改变,逆转录-定量聚合酶链反应(RT-qPCR)检测回肠组织JAK2 mRNA、STAT3mRNA表达,蛋白免疫印迹(Western Blot)法检测回肠组织p-JAK2蛋白、p-STAT3蛋白表达。【结果】SAP大鼠肠组织病理损害严重,JAK2 mRNA、STAT3 m RNA、p-JAK2蛋白和p-STAT3蛋白过度表达;大承气汤、JAK2抑制剂和STAT3抑制剂可明显保护肠道结构,抑制JAK2 m RNA、STAT3 m RNA、p-JAK2蛋白和p-STAT3蛋白过度表达。【结论】活化的JAK2/STAT3通路在SAP肠损害中起重要作用。大承气汤可能通过抑制活化的JAK2/STAT3通路保护SAP大鼠肠道。Objective To explore the protective effect of Dachengqi Decoction on intestinal tract in rats with severe acute pancreatitis(SAP)via Janus kinase 2(JAK2)/signal transducers and transcription 3(STAT3)signaling pathway.Methods SAP rat model was established by retrograde pancreatobiliary tract injection of sodium taurocholate of 3.5%.The successful modeling rats were randomly divided into model group,JAK2 inhibitor group(giving intragastric administration of Ruxolitinib 2 h before operation),STAT3 inhibitor group(giving intraperitoneal injection of Statticb 2 h before operation),Dachengqi Decoction group(giving intragastric administration of Dachengqi Decoction 2 h before operation),additionally,the sham operation group(giving intraperitoneal injection tract injection of saline water)was set up.And then the various groups were further divided into 3-,6-,12-and 18-h subgroups.The pathological changes in terminal ileum were observed by haematoxylin-eosin(HE)staining,the mRNA expression levels of JAK2 and STAT3 were determined by reverse transcription-quantitative polymerase chain reaction(RT-qPCR),and the protein expression levels of p-JAK2 and p-STAT3 in ileum were detected by Western blotting method.Results The SAP rats had severe pathological damage in ileum,and over-expression of JAK2 mRNA,STAT3 mRNA,p-JAK2 protein and p-STAT3 protein in the terminal ileum,while Dachengqi Decoction,JAK2 inhibitor and STAT3 inhibitor could obviously ameliorate these events.Conclusion The activated JAK2/STAT3 pathway plays an important role in SAP intestinal injury.Dachengqi Decoction can protect the intestinal tract possible by inhibiting the activated JAK2/STAT3 pathway.
