管灸对面神经损伤模型家兔Grb2、K-Ras表达的影响

摘  要:目的:探讨管灸促损伤面神经修复的胞内信号转导机制。方法:建立家兔面神经压榨损伤模型,通过HE染色切片观察治疗后各组面神经组织形态改变,并运用ELISA技术检测各组面神经Grb2、K-Ras蛋白的表达。结果:①面神经组织形态改变:管灸组面神经仍可见少量空泡性变,神经纤维直径变细,髓鞘较正常变薄,但程度低于模型组和热疗组,且热疗组与模型组无明显差异;②Grb2、K-Ras蛋白表达:与模型组相比,管灸组Grb2、K-Ras蛋白表达升高,有统计学差异(P<0.05);热疗组指标均无统计学差异(P>0.05)。结论:管灸能上调面神经损伤家兔Grb2、K-Ras蛋白表达,增强NT-Trk信号在胞内转导的MAPK/ERK1/2途径,以促进损伤面神经的修复再生。Objective:To reveal the intracellular signal transduction mechanism of Guan-moxibustion in promoting the re-pair of facial neuritis.Methods:A model of facial nerve injury in rabbits was established.HE staining was used to observe the morphological changes of facial nerve tissue in each group,and the expression of Grb2protein and K-Ras protein in facial nerve of each group were examined individually by ELISA.Results:①The morphological changes of facial nerve tissue:In the Guan-moxibustion group,a small amount of vacuolation was still observed,the diameter of nerve fibers became thinner,and the pulp pin was thinner than normal,but the degree was lower than that of the model group and hyperthermia group.②Expression of Grb2and K-Ras proteins:Comparing with the model group,the protein expression of Grb2and K-Ras of the Guan-moxibustion group was increased(P<0.05).There was no statistical significance in the indexes in the hyperthermia group(P>0.05).Con-clusion:Guan-moxibustion can up-regulate the expression of Grb2and K-Ras protein in facial nerve of model rabbits,thus en-hancing the MAPK/ERK1/2 pathway of NT-Trk signal transduction in cells,and promoting the repair and regeneration of injured facial nerve.
