摘 要:目的:分析针刺对脑卒中模型大鼠认知功能障碍影响及其与磷酸酰肌醇-3-激酶(PI3K)/蛋白激酶B(Akt)信号转导通路联系。方法:选取SPF级雄性大鼠30只,随机数字表法将大鼠分成3组,分别为针刺组、模型组和正常组,针刺组、模型组大鼠制备大脑中动脉梗塞(MCAO)模型,成功造模后电针仪对针刺组大鼠行针刺治疗,模型组与正常组不进行任何处理。观察大鼠神经功能评分、脑梗死面积、脑组织细胞凋亡率、脑组织PI3K、Bcl-2、p-Akt、t-Akt、Bax蛋白表达及Bax、Bcl-2及Bc1-2/Bax mRNA表达。结果:和正常组相比,模型组大鼠脑梗死面积增大;和模型组相比,针刺组大鼠脑梗死面积减少,差异有统计学意义(P<0.05);和正常组相比,模型组大鼠凋亡阳性细胞比例升高;和模型组相比,针刺组大鼠凋亡阳性细胞比例降低,差异均有统计学意义(P<0.05);和正常组相比,模型组大鼠Bax蛋白表达升高,PI3K、Bcl-2、p-Akt蛋白表达降低;和模型组相比,针刺组大鼠Bax蛋白表达降低,PI3K、Bcl-2、p-Akt蛋白表达升高,差异有统计学意义(P<0.05);和正常组相比,模型组大鼠Bax mRNA表达升高,Bcl-2及Bc1-2/Bax mRNA表达降低;和模型组相比,针刺组大鼠Bax mRNA表达降低,Bcl-2及Bc1-2/Bax mRNA表达升高,差异有统计学意义(P<0.05)。结论:针刺脑卒中大鼠可显著改善其认知功能障碍,加速神经功能恢复,作用机制可能将PI3K/Akt路径激活,对神经细胞凋亡抑制有联系。Objective:To analyze the effect of acupuncture on cognitive dysfunction for rats with stroke and its association with Phosphatidylinositol-3-Kinase(PI3K)/protein Kinase B(Akt)signal transduction pathway.Methods:Thirty male SPF rats were randomly divided into three groups:acupuncture group,model group and normal group.The acupuncture group and the model group were used to prepare middle cerebral artery infarction(MCAO)model.After successful modeling,the electroa-cupuncture instrument was treated with acupuncture in the acupuncture group,and the model group and the normal group were not treated.The neurological function score,cerebral infarction area,brain tissue apoptosis rate,brain tissue PI3K,Bcl-2,p-Akt,t-Akt,Bax protein expression and Bax,Bcl-2and Bcl-2/Bax mRNA were observed.Results:Compared with the nor-mal group,the infarct size of the model group increased.Compared with the model group,the infarct size of the rats in the acu-puncture group decreased,the difference was statistically significant(P<0.05)compared with the normal group.The proportion of apoptosis-positive cells in the model group was increased.Compared with the model group,the percentage of apoptosis-posi-tive cells in the acupuncture group was decreased,and the difference was statistically significant(P<0.05).Compared with the normal group,the expression of Bax protein of the model group was increased and the expression of PI3K,Bcl-2and p-Akt pro-tein was decreased in the rats.Compared with the model group,the expression of Bax protein was decreased and the expression of PI3K,Bcl-2and p-Akt protein was increased in the acupuncture group.The difference was statistically significant(P<0.05).Compared with the normal group,the expression of Bax mRNA was increased and the expression of Bcl-2and Bcl-2/Bax mRNA was decreased in the model group.Compared with the model group,acupuncture was performed.The expression of Bax mRNA was decreased and the expression of Bcl-2and Bcl-2/Bax mRNA was increased in the rats.The difference was statistically signific
