摘  要:目的:探究银杏叶提取物对心肌缺血再灌注损伤(MI/RI)大鼠心肌功能的保护作用。方法:50只SD大鼠随机分为假手术组(Sham组)、模型组(Mod组)、银杏叶提取物高剂量组(GBE-H组,600mg/kg),银杏叶提取物剂量组(GBE-L组,300mg/kg),每组10只。连续灌胃给药7天后建立大鼠MI/RI模型。测量各组大鼠舒张末期左室内径(LVIDd)、收缩末期左室内径(LVIDs)、短轴缩短率(FS)、射血分数(EF)、每搏输出量(SV);计算心脏/体质量比及心肌梗死面积;检测血清心肌酶[谷草转氨酶(AST)、磷酸肌酸激酶(CPK)、乳酸脱氢酶(LDH)]含量;计算凋亡指数(AI);测定心肌组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性及丙二醛(MDA)含量。结果:与Sham组比较,Mod组大鼠LVIDd及LVIDs升高,FS、EF及SV降低(P均<0.05);AI、心脏/体质量比、心肌梗死面积明显增大(P<0.05);血清心肌酶含量明显升高(P<0.05);心肌组织SOD及CAT活性降低,MDA含量升高(P均<0.05)。与Mod组比较,GBE-H组及GBE-L组大鼠LVIDd及LVIDs降低,FS、EF及SV升高(P均<0.05);AI、心脏/体质量比、心肌梗死面积明显减小(P<0.05);血清心肌酶含量均明显降低(P<0.05);心肌组织SOD及CAT活性升高,MDA含量降低(P均<0.05)。结论:银杏叶提取物对MI/RI大鼠的心肌功能有显著的保护作用,其机制可能与银杏叶提取物提高氧自由基清除能力,降低脂质过氧化损伤,减轻心肌梗死程度有关。Objective:To explore the protective effect of Ginkgo Biloba Extract on myocardial function for rats with myocar-dial ischemia-reperfusion injury(MI/RI).Methods:50SD rats were randomly divided into sham operation group(Sham group),model group(Mod group),high-dose Ginkgo Biloba Extract group(GBE-H group,600mg/kg)and low-dose Ginkgo Biloba Extract(GBE-L group,300mg/kg),with 10rats in each group.Rat MI/RI model was established after 7d of continuous intragastric administration.The left ventricular internal diameter at end-diastole(LVIDd),left ventricular internal diameter at end-systole(LVIDs),fractional shortening(FS),ejection fraction(EF)and stroke volume(SV)were measured.The heart/body mass ratio and myocardial infarct area were calculated.The counts of serum myocardial enzymes[aspartate aminotransferase(AST),creatine phosphate kinase(CPK),lactate dehydrogenase(LDH)]were detected.The apoptotic index(AI)was cal-culated.The superoxide dismutase(SOD),catalase(CAT)activity and malondialdehyde(MDA)content in myocardial tissues were measured.Results:Compared with the Sham group,LVIDd and LVIDs were increased while FS,EF and SV were de-creased in the Mod group(P<0.05).AI,heart/body mass ratio and myocardial infarct area were significantly increased(P<0.05).Serum myocardial enzymes contents were increased significantly(P<0.05).SOD and CAT activity in myocardial tissues were decreased while MDA content was increased(all P<0.05).Compared with the Mod group,LVIDd and LVIDs were de-creased while FS,EF and SV were increased in GBE-H group and GBE-L group(P<0.05).AI,heart/body mass ratio and myocardial infarct area were significantly decreased(P<0.05).Serum myocardial enzymes contents were significantly decreased(P<0.05).SOD and CAT activity in myocardial tissues were increased while MDA content was decreased(P<0.05).Conclu-sion:Ginkgo Biloba Extract has significant protective effect on myocardial function in MI/RI rats.Its mechanism may be related to the improvement of oxygen free radical scavenging ability,reduction of lipid
