摘 要:目的探讨使用链脲霉素诱导母系的糖尿病模型后子代新生儿期大鼠脂肪组织的代谢变化。方法选取雌性SB大鼠20只,使用链脲霉素诱导雌性大鼠成为糖尿病模型,成年后与正常雄性大鼠交配,将雄性子代幼鼠按照母系是否患有糖尿病分为实验组(20只)及对照组(10只),雄性子代大鼠于12周时处死并获取附睾及皮下脂肪垫,提取脂肪细胞评估基础水平及胰岛素诱导的2-脱氧葡萄糖(2DG)的摄取,葡萄糖氧化为CO2及结合葡萄糖成为脂质、脂肪分解的能力。结果与对照组相比较,实验组大鼠体重、附睾脂肪垫重量及平均脂肪直径均增加(P<0.05)。两组皮下脂肪垫的脂肪细胞直径及脂肪垫重量、葡萄糖耐量及胰岛素水平两组比较差异均无统计学意义(P>0.05)。实验组附睾脂肪细胞基础及胰岛素诱导下葡萄糖摄取量较对照组增加(P<0.05),同时实验组葡萄糖氧化为CO2及转为脂质增加(P<0.05),但体外脂质分解的能力无明显改变(P>0.05)。与对照组比较,GLUT4、胰岛素受体(IRβ)及乙酰辅酶A(ACC)等在实验组脂肪垫中均增加(P<0.05)。结论大鼠母系糖尿病可对子代的脂肪代谢产生影响,脂质储存能力增加但脂质分解的能力没有明显改变。Objective o investigate the metabolic changes of adipose tissue in neonatal rats after the induction of maternal diabetes model with streptozotocin.Methods 20 female SD rats were selected and induced into diabetic model by streptozotocin.The male offspring rats were divided into experimental group(20 rats)and control group(10 rats)according to whether the female had diabetes or not.The male offspring rats were killed at 12 weeks and the epididymis and subcutaneous fat pad were obtained.The basic level of fat cell evaluation and islets of Langerhans were extracted The uptake of 2-deoxyglucose(2DG),the ability of glucose to oxidize to CO2 and combine with glucose to form lipid and fat.Results Compared with the control group,the body weight,the weight of epididymal fat pad and the average diameter of fat in the experimental group increased(P<0.05).There was no significant difference in the diameter and weight of fat cells,glucose tolerance and insulin level between the two groups(P>0.05).Compared with the control group,the basal cells of epididymal fat cells and insulin-induced glucose uptake in the experimental group increased(P<0.05),while the oxidation of glucose to CO2 and its conversion to lipid increased(P<0.05),but the ability of lipid decomposition in vitro had no significant change(P>0.05).Compared with the control group,GLUT4,insulin receptor(IRβ)and acetyl CoA(ACC)were increased in the fat pad of the experimental group(P<0.05).Conclusion Maternal diabetes mellitus can affect the fat metabolism of offspring.The ability of lipid storage increases,but the ability of lipid decomposition does not change significantly.
