摘 要:为优化乌骨藤(Marsdenia tenacissima)中白桦脂酸的提取工艺,在单因素实验的基础上,采用Box-Behnken中心组合设计方法,研究超声功率、提取时间、料液比及提取次数等因素及其交互作用对白桦脂酸提取率的影响。利用Design Expert 8.06软件对数据进行回归分析,得到二次多项式回归方程的预测模型,并通过响应曲面优化方法得到白桦脂酸最佳提取工艺。结果表明,乌骨藤中白桦脂酸的最佳提取工艺为超声功率150 W,提取时间20 min,料液比(g/mL)为1:25,提取2次,白桦脂酸提取率预测值为0.314%,验证值为0.311%。Response surface methodology was used to optimize the extraction technology of betulinic acid from Marsdenia tenacissima.Based on single-factor experiments,ultrasonic power,extraction duration,material-liquid ratio,and times of extraction were selected as influencing factors during extraction.The experiment mathematical model was arranged according to Box-Behnken central composite experiment design,which studied each variable and their interactions on the extraction yield of betulinic acid.The quadratic multinomial model on the extraction yield of betulinic acid and each influence factor of extraction betulinic acid from M.tenacissima was established by using the Design-Expert software.And the optimum conditions were as follows:ultrasonic power 150 W,material-liquid ratio 1:25,extraction duration 20 min and extraction twice.The predicted value of extraction yield of betulinic acid was 0.314%under the optimum conditions,and the experimental value was 0.311%.
