摘 要:目的验证长链非编码RNA(lncRNA)中的分化拮抗非编码RNA(DANCR)在常见恶性肿瘤中的作用和预后价值。方法检索PubMed、Web of Science、知网和万方数据库中关于lncRNA DANCR表达与人类癌症关系的相关文献,检索时间为建库至2019年11月1日。根据系统综述和荟萃分析优先报告的条目声明进行文献的筛选和数据提取,运用Stata 12.0软件分析比值比(OR)和相应的95%置信区间(CI)来评估癌症患者肿瘤浸润深度、淋巴结转移、远处转移、肿瘤分化程度、肿瘤浸润深度的促进或抑制效应。在临床分期的总体生存率上,计算风险比(HR)和相应的95%CI来评估生存期的效应。结果共纳入10项研究,包括2143例癌症患者。与DANCR低表达组患者相比,DANCR高表达组患者的总生存期较短(HR=1.26,95%CI 1.18~1.34,P<0.001)。消化道肿瘤患者DANCR高表达组的总生存期短于DANCR低表达组(HR=1.20,95%CI 1.12~1.30,P<0.001);呼吸系统肿瘤DANCR高表达组的生存期短于DANCR低表达组(HR=1.43,95%CI 1.24~1.64,P<0.001)。肿瘤分化程度(OR=2.72,95%CI 1.01~7.29,P=0.047)、肿瘤浸润深度(OR=1.62,95%CI 1.13~2.31,P=0.009)与DANCR高表达有关,lncRNA DANCR高表达患者的肿瘤分化程度更差,肿瘤浸润深度更深。结论lncRNA DANCR升高可作为预测多种癌症预后不良的潜在生物标志物,但需要纳入更多新的研究做进一步的分析。Objective To verify the role and prognostic value of differentiation antagonistic non-coding RNA(DANCR)in long-chain non-coding RNA(lncRNA)in common malignant tumors.Methods We searched PubMed,Web of Science,CNKI and Wanfang database for research articles on the relationship between the expression of lncRNA DANCR and human cancer,from database establishment till Nov.1st,2019.Literature screening and data extraction were carried out according to the statement of preferred reporting items for systematic reviews and Meta-analyses.The odds ratio(OR)and 95%confidence interval(CI)were calculated and analyzed by Stata 12.0 software to evaluate the tumor invasion depth,lymph node metastasis,distant metastasis,tumor differentiation and tumor invasion depth promotion or inhibition effects.In the overall survival rate of clinical stages,the hazard ratio(HR)and the corresponding 95%CI were calculated to evaluate the effect of survival.Results Altogether 10 studies were included,involving 2143 cancer patients.Compared with patients in the DANCR low-expression group,patients in the DANCR high-expression group had shorter overall survival(HR=1.26,95%CI 1.18-1.34,P<0.001).The overall survival time of patients with gastrointestinal tumors in the DANCR high-expression group was shorter than that in the DANCR low-expression group(HR=1.20,95%CI 1.12-1.30,P<0.001);the survival time of respiratory tumor in the DANCR high-expression group was shorter than that in the DANCR low-expression group(HR=1.43,95%CI 1.24-1.64,P<0.001).The degree of tumor differentiation(OR=2.72,95%CI 1.01-7.29,P=0.047)and the depth of tumor invasion(OR=1.62,95%CI 1.13-2.31,P=0.009)were related to high expression of DANCR,and the tumors of the patients with high expression of lncRNA DANCR had worse differentiation and deeper tumor invasion.Conclusion The increase of lncRNA DANCR can be used as a potential biomarker to predict the poor prognosis of various cancers,but more updated studies need to be included for further analysis.
