摘 要:目的探讨孕期香烟烟雾暴露(PCSE)对雄性子代大鼠生殖功能的影响。方法大鼠在孕14-18天给予香烟烟雾暴露30 min/d,一部分孕鼠于孕20天剖宫产取雄性胎鼠;另一部分孕鼠自然分娩,其子代雄性于90日龄处死。称重孕鼠和仔鼠体重,并分别检测胎鼠和仔鼠的血性激素含量、睾丸组织甾体合成酶的基因水平、睾丸间质细胞(LCs)数量和成年子代附睾精子数量和活力。结果与对照组相比,PCSE组胎血睾酮(T)降低(P<0.01),胎睾丸LCs数量(P<0.05)和类固醇生成急性调节蛋白(Star)、3β-羟类固醇脱氢酶(3β-hsd)的基因表达均降低(P<0.05)。PCSE组成年血睾酮降低,黄体生成素(LH)及LH:T升高(P<0.05),睾丸LCs数量和Star、3β-hsd的基因表达降低,附睾尾精子数量和活力降低(P<0.05)。结论PCSE可致子代宫内时期血睾酮水平降低,可延续至出生后成年,并伴随成年时期精子数量的减少和质量的下降,睾丸LCs减少和甾体酶Star及3β-hsd的表达降低可能是其主要机制之一。Objective To explore and discuss the influence of prenatal cigarette smoke exposure(PCSE)on reproductive function in offspring rats.Methods The pregnant rats were exposed to cigarette smoke for 30 min/d on gestation day 14-18.Male fetal rats were taken from some pregnant rats through the caesarean on gestation day 20,and the rest pregnant rats gave natural childbirth.The male offspring rats were killed on postnatal day 90.The pregnant and offspring rats were weighed.The levels of serum sex hormones,the gene level of testicular steroid synthase,and the count of Leydig cells(LCs)of fetal and offspring rats were detected and measured.Furthermore,the epididymal sperm count and vitality of adult offsprings weremeasured.Results Compared with the control group(0.84+0.02 ng/ml),the content of testosterone in the fetal serum(0.53+0.03 ng/ml)in the PCSE group significantly decreased(P<0.01).The fetal testicular LCs counts decreased(P<0.05),the mRNA expression of steroidogenic acute regulatory protein(Star)andthe 3β-hydroxysteroid dehydrogenase(3β-hsd)in fetal testis in PCSE group decreased compared with that of the control group(P<0.05).In adult rats,compared with the control group,the content of the serum testosterone in the PCSE group decreased(P<0.05),the serum luteinizing hormone(LH)increased(P<0.05),the ratio of LH to T increased(P<0.05),and testicular LCs counts was lower.Meanwhile,the m RNA expression of testicular Star and 3β-hsd of adult offspring rats in PCSE decreased(P<0.05).Furthermore,the epididymal sperm count and vitality in PCSE were lower than that of the control group(P<0.05).Conclusion PCSE resulted in the low serum testosterone in the intrauterine period,which could be extended to the postnatal adulthood with the decrease of the sperm count and vitality.The decreased testicular LCs count and the low expression of Star and 3β-hsd caused by PCSE may be one manifestation of the primary mechanisms.
