枣仁天麻胶囊食用安全性及其对小鼠睡眠的影响

摘　　要：目的探讨枣仁天麻胶囊食用安全性及其对小鼠睡眠的影响。方法ICR小鼠行急性毒性实验及3项遗传毒性实验(Ames实验、骨髓微核实验和精子畸形实验)。低、中、高剂量组SD大鼠采用枣仁天麻胶囊灌胃,每日670、1340、2680 mg/kg,空白对照组灌无菌水,每组20只,连续给药30 d后检测血液学指标及血生化指标并进行组织病理学检查。120只CI/F1代小鼠均分为四组,低、中、高剂量组分别采用枣仁天麻胶囊每日130、270、800 mg/kg灌胃,空白对照组灌无菌水,行睡眠实验。结果连续喂饲大鼠30 d后,低、中、高剂量组血常规及血生化指标与空白对照组比较差异均无统计学意义(P>0.05)。小鼠枣仁天麻胶囊急性经口最大耐受量>15000 mg/kg,属无毒级。3项遗传毒性实验结果均为阴性。高剂量组小鼠睡眠时间长于空白对照组,且睡眠潜伏期短于空白对照组(P<0.05)。结论枣仁天麻胶囊可能改善了小鼠睡眠,有一定的疗效。Objective To explore the food safety of Zaoren Tianma capsule(ZTC)and its effect on sleep in mice.Methods Acute toxicity test and three genotoxicity tests(Ames test,bone marrow micronucleus test and sperm deformity test)were performed in 20 ICR mice.The SD rats were divided into 4 groups and were given ZTC 670,1340,2680 mg/kg and water daily by gastric gavage,respectively.The hematological and biochemical indicators were detected and histopathological examinations were carried out after gastric gavage for continuous 30 days.A total of 120 CI/F1 mice for sleep experiments was equally divided into 4 groups and ZTC 130(group A),270(group B)and 800 mg/kg(group C)were given by gastric gavage daily for continuous 30 days.Group D was taken as the blank control.Results There was no significant difference in blood routine and blood biochemical indexes between the rats took ZTC and those in blank control group(P>0.05).Acute toxicity test showed that the maximum tolerant dosage of ZTC in mice was more than 15000 mg/kg,which was considered as nontoxic rank.The results of three genotoxicity tests were all negative.Compared to group D,group A had a longer sleep time and shorter sleep latency(P<0.05).Conclusion ZTC can improve mice sleep with good edible safety.
