摘  要:过量表达内源EPSPS基因不仅能够提高栽培稻对草甘膦除草剂的抗性,还能在不施用草甘膦的条件下提高转基因栽培稻与杂草稻杂种后代的光合速率.由于叶绿素是影响光合作用的关键因子,本研究探索了过量表达EPSPS基因是否提高栽培稻与杂草稻杂种后代的叶绿素含量以及叶绿素合成途径中关键基因的表达.以EPSPS转基因栽培稻(EP3)及其与杂草稻两个组合的F3杂种后代分离群体为实验材料,分别测定了含EPSPS转基因和不含EPSPS转基因杂种后代群体的叶绿素含量以及叶绿素合成关键基因的表达量.结果表明,含转基因EPSPS的栽培稻×杂草稻后代群体的叶绿素含量显著增高,在培养至30d的时候,总叶绿素含量分别增高了86.89%和22.58%;培养至60d的时候,总叶绿素含量分别增高了72.82%和51.30%.叶绿素合成途径中的3个关键基因GLUTR、CHLH和PORA的表达显著上调.因此而做出如下结论,EPSPS基因的过量表达可以通过调控叶绿素合成途径中关键基因的表达,促进叶绿素的合成,进而提高了植物光合作用的速率.Overexpression of EPSPS in rice usually increases glyphosate herbicide tolerance,but previous studies indicated that overexpression of EPSPS increased rates of the photosynthesis in hybrid progeny(F3)between cultivated rice and weedy rice.As a key component,chlorophyll plays a key role in photosynthesis,therefore,investigating the reason why overexpression of transgenic EPSPS increased the photosynthetic rate of the hybrid progeny is very important.We studied chlorophyll contents and expression of the key genes(GLUTR,CHLH and PORA)that are related to chlorophyll synthesis,using transgenic(WH++)and non-transgenic(WH--)lineages derived from hybrids between cultivated rice with EPSPS transgene(EP3)and two weedy rice populations.Results indicated that the total chlorophyll contents of the two transgenic hybrid lineages significantly increased by 86.89%and 22.58%,respectively,compared to their non-transgenic controls at 30 days;while those increased significantly by 72.82%and 51.30%at 60 days.In addition,results showed that expression of the three key genes(GLUTR,CHLH and PORA)was also significantly increased.It is therefore concluded that overexpression of the EPSPS gene can promote chlorophyll synthesis by upregulating expression of the key genes in the chlorophyll synthesis pathway and enhance photosynthesis in plants.
