摘 要:【目的】PHO2是磷信号通路中重要的调控因子,PHO2突变会造成植物过量的磷累积。尽管已对PHO2进行了大量研究,但始终聚焦于磷元素的积累上,PHO2是否对其他元素的积累有影响尚不清楚。【方法】本研究首先利用水稻日本晴T-DNA插入突变体ospho2,研究在该突变背景下矿质元素在植株体内累积的情况。因Na在水稻和高粱pho2突变体种子中大量累积,利用水稻和高粱pho2突变体同时进行种子萌发期耐盐胁迫实验。【结果】分析野生型日本晴和ospho2突变体在成熟期时地上部和种子中大量元素和微量元素的含量,表明在地上部中ospho2突变体植株中不仅超量累积了磷元素,还累积了诸如钾、镁、锰等元素;在ospho2突变体种子中也过量累积了磷元素,但是累积程度远不如地上部分,其他元素钾、镁、钠、钙和锌等也都有一定的累积。盐胁迫下水稻和高粱pho2突变体萌发期的芽苗率较野生型显著降低。【结论】OsPHO2的突变不仅导致磷的超量累积,对其他很多元素的积累也有较大的影响。水稻和高粱pho2突变体降低了种子萌发期耐盐性。【Objective】Phosphorus is one of the three important minerals and plays an important role in plant growth and development.Molecular signaling pathways such as uptake,transportation and utilization of phosphorus have been well studied in Arabidopsis and rice.OsPHO2 gene is one of key regulatory factors in rice phosphorus signaling pathway.Mutations in Arabidopsis or rice PHO2 gene causes excessive Pi accumulation in mutant plants.Despite a large number of studies on the PHO2 gene,it has always been focused on the Pi accumulation.It is not clear whether the PHO2 gene affects other non-Pi minerals accumulation.【Method】In the present study,a rice Nipponbare T-DNA insertion mutant ospho2 was used to analyze the accumulation of P and non-P elements.Because of overaccumulation of Na in rice and sorghum mutant pho2 seeds,we used rice and sorghum pho2 mutants to analyze salt tolerance during seed germination.【Result】We analyzed the contents of macroelement and microelement in shoots and seeds of wild-type Nipponbare and ospho2 mutant at maturity stage.Results showed that phosphorus was not the only one element accumulated in shoots of ospho2 mutant plants,but potassium,magnesium,manganese and other elements also accumulated in the mutant;phosphorus also accumulated excessively in the seeds of ospho2 mutant,but the accumulation ratio was far less than the shoots,and other elements such as potassium,magnesium,sodium,calcium and zinc also accumulated in the seeds of ospho2.Seeds germination and seedlings survival rates of rice and sorghum plants under 1.2%NaCl treatment showed reduced performance compare with WT.【Conclusion】Our study shows that mutation of OsPHO2 leads not only excessive accumulation of phosphorus,but also has a great influence on accumulation of several other non-P elements.Rice and sorghum pho2 mutants reduced salt tolerance during seed germination.
