摘 要:【目的】研究菌草灵芝多糖肽(GL-PP)对肝癌细胞Huh7活性、迁移和细胞凋亡的影响,为GL-PP应用于肝癌治疗提供理论依据。【方法】用低(50μg/mL)、中(100μg/mL)、高(200μg/mL)3种不同浓度的GL-PP处理肝癌细胞Huh7,分别采用CCK8、Transwell方法、流式细胞仪分析不同浓度GL-PP对肝癌细胞Huh7活性、迁移和细胞凋亡的影响。【结果】培养基添加GL-PP后肝癌细胞Huh7活力降低,且GL-PP添加浓度越高活力降低越多,但是差异均不显著(P>0.05);GL-PP的浓度为100μg/mL时对肝癌细胞Huh7的迁移无明显影响,当浓度为200μg/mL时对肝癌细胞Huh7的迁移具有明显的抑制作用;低浓度(50μg/mL)和中浓度(100μg/mL)的GL-PP对肝癌细胞Huh7的凋亡无明显影响(P>0.05),高浓度(200μg/mL)的GL-PP能够明显(P<0.05)诱导肝癌细胞Huh7的凋亡。【结论】GL-PP对肝癌细胞Huh7细胞活性影响不显著,高浓度的GL-PP可抑制肝癌细胞Huh7的迁移,也可诱导其凋亡。【Objective】The present paper aimed to study the effects of Ganoderma lucidum polysaccharide peptide(GL-PP)on hepatocarcinima cells Huh7 activity,migration and apoptosis which provides a theoretical basis for the application of GL-PP in the treatment of liver cancer.【Method】Hepatocarcinima cells Huh7 were treated with low concentration(50μg/mL),medium concentration(100μg/mL)and high concentration(200μg/mL)of GL-PP,followed by CCK8,Transwell and flow cytometry method to analyze the effects of different concentrations of GL-PP on Huh7 activity,migration and apoptosis,respectively.【Result】The Huh7 cell viability decreased after adding GL-PP in the medium.Furthermore,the cell viability decreased with the increasing concentration of GL-PP but not significantly different(P>0.05).The migration of Huh7 cells was not significantly affected when the concentration of GL-PP was 100μg/mL.However,the migration of Huh7 cells was significantly inhibited as the concentration of GL-PP was 200μg/mL.Low concentration(50μg/mL)and medium concentration(100μg/mL)of GL-PP had no significant effect on the apoptosis of hepatocarcinima cells Huh7(P>0.05),while high concentration(200μg/mL)of GL-PP could significantly(P<0.05)induce apoptosis of Huh7 cells.【Conclusion】GL-PP had no significant effect on the activity of Huh7 cells,but high concentration of GL-PP could inhibit the migration of Huh7 cells and induce apoptotic process.
