摘 要:该文将具有高电导率的金纳米颗粒@共价有机框架(Au NPs@COF-LZU8)作为基底固定CRP抗体,构建了一种检测迅速、灵敏度高的无标记型电化学免疫传感器用于实际血清样品中C-反应蛋白(CRP)的测定。当抗体与抗原发生免疫反应时,形成的免疫复合物会阻碍电化学探针[Fe(CN)6]^4-/3-的电子传递,降低其响应电流,从而实现CRP的快速检测。此外,Au NPs@COF-LZU8具有丰富的吸附位点和较大的比表面积,可以通过提高抗体的负载量来改善免疫传感器的性能。在最优实验条件下,此电化学免疫传感器的线性范围从0.1 ng/mL^200 ng/mL,相关系数R2=0.9913,检出限为33.3 pg/mL,将此免疫传感器用于人血清样品中CRP的检测,结果令人满意。此策略也可用于构建其它免疫传感器。A fast, sensitive, label-free electrochemical immunosensor was developed to measure C-reactive protein(CRP) in actual serum samples using Au NPs@COF-LZU8 with high conductivity as a substrate to immobilize CRP antibodies has. When CRP binds with CRP antibody, the electric signal of Fe(CN)6^4-/3-was obstructed by the formed immuno complex, and the detection of CRP can be realized. In addition, Au NPs@COF-LZU8 has rich adsorption site and large specific surface area, which can greatly increase the loading amount of antibody and significantly improve the performance of the immunosensor. Under the optimal experimental conditions, the linear range of the electrochemical immunosensor ranged from 0.1 ng/mL to 200 ng/mL, the correlation coefficient was 0.9913 with the detection limit of 33.3 pg/mL. The immunosensor was used to detect CRP in human serum samples. The result is satisfactory. This strategy can also be used to construct other immunosensors.
