摘 要:致病疫霉释放的游动孢子是其侵染植物并引起病害的主要因子之一。为快速获得大量侵染力强的游动孢子,并明确拮抗菌HT-6对游动孢子活性的影响,首先将经黑麦培养基(RA)培养获得的致病疫霉孢子囊分别用自来水(TW)、无菌蒸馏水(SDW)、10%V8培养液(V8)和黑麦液体培养基(RL)处理,比较游动孢子的释放能力、侵染能力、运动能力和成膜能力的差异,在此基础上检测不同浓度HT-6菌液对来自V8的游动孢子活性的影响。结果显示,不同培养液处理后,游动孢子释放率在TW中最高(46.77%),在RL中最低(6.16%),但来自V8的游动孢子对马铃薯块茎切片的侵染能力(病情指数为87.51)、运动能力(23.25%)和成膜能力(OD540=1.107)均为最强;以不同浓度HT-6菌液处理来自V8的游动孢子后,发现随菌液浓度升高,细菌HT-6对游动孢子活性的抑制作用逐渐增强,在OD600为1.433浓度时游动孢子侵染块茎切片的病情指数为23.55、运动能力为2.97%,成膜能力被完全抑制。这些结果表明,利用V8制备致病疫霉游动孢子显著优于RL、SDW及TW,且细菌HT-6可以强烈抑制游动孢子的活性。Zoospores produced by Phytophthora infestans are one of the main factors that infect plants and cause plant diseases. In order to quickly obtain a large number of zoospores with strong infectivity, and to explore the effect of antagonistic bacterium HT-6 on zoospore activity, sporangia obtained from rye medium(RA) were treated with tap water(TW), sterile distilled water(SDW), 10% V8 culture medium(V8) and rye liquid medium(RL) to compare the release ability, infection ability, motile ability, and biofilm-forming ability of zoospores, and further to investigate the effects of different concentrations of HT-6 bacterial suspension on activity of zoospore produced in V8 treatment. The results showed that, after treatment with different culture media, zoospore release rate was the highest in TW(46.77%) and the lowest in RL(6.16%), but the strongest infection ability on potato tuber slices(disease index 87.51), motile ability(23.25%), and biofilm-forming ability(OD540=1.107) of zoospores were found in V8 treatment. After treating zoospores from V8 with different concentrations of HT-6 bacterial suspension, the inhibition of bacterial suspension on zoospore activity was enhanced as the bacterial suspension concentrations increased, and when the OD600 was 1.433, the diseases index on tuber slices and motile ability were 23.55 and 2.97%, respectively, and the biofilmforming ability was completely inhibited. These results indicate that zoospores of P. infestans prepared by V8 is significantly superior to RL, SDW, and TW, and the bacteria HT-6 can strongly inhibit the activity of zoospores.
