摘  要:目的以弓形虫强毒株感染小鼠为模型研究,从发病情况、生存时间、小鼠体重变化、肌酐水平探究白藜芦醇对急性弓形虫病的治疗效果及毒副作用,同时初步探究白藜芦醇在此模型中的作用机制。方法将20只小鼠按体重均匀分成溶剂组、白藜芦醇组、乙胺嘧啶组、联合用药组,每组5只,分别予以0.5 mL羧甲基纤维素(溶剂)、白藜芦醇悬液、乙胺嘧啶悬液、白藜芦醇+乙胺嘧啶悬液灌胃2 d后,通过腹腔注射染虫并继续灌胃5 d,记录小鼠的体重与生存情况。按上述方法对小鼠进行灌胃感染后7 d处死,取血清检测小鼠肌酐水平并提取原代腹腔巨噬细胞检测沉默信息调节因子2相关酶1(SIRT1)蛋白的表达。结果白藜芦醇可以减缓由乙胺嘧啶毒副作用造成的体重减轻和血清肌酐升高,且差异有统计学意义(P<0.05),同时上调体内SIRT1表达。但白藜芦醇单剂及联合乙胺嘧啶用药均不能延长小鼠生存时间,差异无统计学意义(P>0.05)。结论白藜芦醇能在一定程度上延缓强毒株弓形虫感染小鼠的发病,并减轻乙胺嘧啶的毒副作用,其治疗效果可能有赖于其对SIRT1蛋白的上调作用。Objective Mice infected with Toxoplasma gondii virulent strain were used to explore the therapeutic and side effects of resveratrol on acute toxoplasmosis from the aspects of survival time,weight change and creatinine level. And the mechanism of resveratrol in this model was explore. Methods Twenty mice were evenly divided into placebo,resveratrol,ethylamine pyrimidine and combination group according to the weight. Each group separately treated with 0.5 mL carboxymethyl cellulose(placebo),resveratrol,ethylamine pyrimidine,resveratrol and ethylamine pyrimidine by gavage. 2 days after treatment mice were infected by intraperitoneal injection,and then keep gavaging for 5 days. Mice weight and survival condition were recorded. Mice were sacrificed 7 days after gastric infection;the level of serum creatinine was determined;the primary abdominal macrophages were colleted for the detection of sirtuin 1(SIRT1)protein by western blot.Results Resveratrol could relieve the weight loss and the increase of serum creatinine caused by the side effects of pyrimethamine(P<0.05),it could also promote the SIRT1 production at the same time. However,neither resveratrol nor combined treatment could help mice survive(P>0.05). Conclusion Resveratrol could delay the morbidity and mortality of mice infected with Toxoplasma gondii. The therapeutic effect might depend on the up-regulation of SIRT1 protein.
