摘 要:以平邑甜茶幼苗为试材,在水培条件下研究了硫酸镉(CdSO4)处理下外源Ca^2+、Ca^2+螯合剂EGTA、质膜Ca^2+通道抑制剂La^3+、钙调素抑制剂CPZ及Ca^2+-ATP酶抑制剂Van对根系活性氧生成和细胞损伤的影响。结果表明:随着0.2mmol·L^-1CdSO4处理时间的延长,根系线粒体膜透性增大、膜电位和Cyt c含量降低、过氧化氢含量、超氧阴离子产生速率、丙二醛含量和细胞死亡数量不断提高,根系发生严重损伤。低浓度的外源Ca^2+(5mmol·L^-1或10mmol·L^-1CaCl2)、EGTA、La^3+和CPZ阻止了根系线粒体膜透性的增大、膜电位和Cyt c含量的下降,抑制了过氧化氢和超氧阴离子的产生,减少了根系细胞死亡量;高浓度的外源Ca^2+(50mmol·L^-1)和Van则加剧了活性氧生成和根系损伤。以上结果显示:钙参与了镉对平邑甜茶根系损伤的诱导,低浓度的CaCl2、EGTA、La3+和CPZ能够缓解CdSO4引起的平邑甜茶幼苗根系线粒体损伤,减少活性氧生成和细胞死亡。Effects of exogenous calcium,Ca^2+-chelate EGTA,Ca^2+channel blocker LaCl3,calmodulin inhibitor CPZ and Ca^2+-ATPase inhibitor Van on the generation of reactive oxygen species(ROS)and cell damage to cadmium stress were investigated in roots of Malus hupehensis Rehd.seedlings grown in hydroponic systems.Results showed that:the content of hydrogen peroxide(H2O2),the production rate of superoxide anion(O2.^-)and the number of cell death in roots were gradually increased with the prolonging of 0.2 mmol•L^-1 CdSO4 treatment.Accumulation of H2O2,production of O2.^-and cell death were significantly inhibited treated with low concentration of exogenous 5 mmol•L^-1 or 10 mmol•L^-1 CaCl2,EGTA,La3+and CPZ under cadmium stress.High concentration of exogenous 50 mmol•L^-1 CaCl2,and Van aggravated the increase of H2O2 content and production rate of O2.-caused by cadmium stress,promoted the cell death in roots.These suggested that calcium was involved in the induction of cadmium damage to roots of Malus hupehensis Rehd.,and low concentration CaCl2,EGTA,La^3+and CPZ could alleviate the generation of ROS and cell death induced by cadmium stress.
