朱氏润肠方对慢传输型便秘大鼠的作用及其机制研究

摘　　要：目的:观察朱氏润肠方对慢传输型便秘(STC)的作用并探讨其机制。方法:将50只大鼠按随机数字表法分为正常组、模型组、朱氏润肠方高剂量组、低剂量组和莫沙必利组。除正常组外,其余大鼠给予大黄灌胃复制STC模型,并给予相应药物干预。记录大鼠24 h粪便总量与粪便含水量,活性炭悬液推进法测定大鼠的肠道传输功能,免疫组化检测大鼠结肠黏膜AQP3、AQP8的表达,RT-PCR检测大鼠结肠AQP3、AQP8 mRNA的表达,Western检测大鼠结肠PI3K、akt的表达。结果:与正常组比较,模型组大鼠24 h粪便总量与粪便含水量明显减少,肠道碳末推进率降低,AQP3、AQP8的表达增高,PI3K、akt的磷酸化水平升高,差异有统计学意义;朱氏润肠方能增加大鼠24h粪便总量与粪便含水量,提高肠道碳末推进率,降低AQP3、AQP8的表达,抑制PI3K、akt的磷酸化水平。结论:朱氏润肠方对STC具有明显的改善作用,其机制可能与抑制PI3K/akt信号通路,从而降低AQP3和AQP8的表达、减少肠道对水的重吸收、增加肠液的分泌有关。Objective:To investigate the effect of Zhushi Runchang decoction on slow transit constipation(STC)in rats,and to clarify its possible mechanism.Methods:The 50 male rats were randomly divided into normal group,model group,ZSRC(28.8 g/kg)group,ZSRC(57.6 g/kg)group and Mosapride group.Rats,except normal group,were given rhubarb to establish the model of STC.After treatment,the 24 hour stool weight and moisture content of feces were recorded,and the carbon powder propulsion in the colon of rats were measured.The expression of AQP3 and AQP8 were detected by immunohistochemistry and Real-time PCR.The expression of PI3K/akt were detected by Western Blotting.Results:Compared with normal group,24 hour stool weight and moisture content of feces in STC rats were decreased;the carbon powder propulsion in the colon of STC rats were also decreased;The expression of AQP3,AQP8 and PI3K/akt were increased.After treatment,Zhushi Runchang decoction can increase 24 hour stool weight,moisture content of feces and carbon powder propulsion,decrease the expression of AQP3,AQP8 and PI3K/akt.Conclusion:Zhushi Runchang decoction have a therapeutic effect on STC,which may be involved with the regulation of absorption and secretion of moisture achieved by depressing AQP3 and AQP8 through PI3K/akt signaling pathway.
