摘 要:目的利用纳米酶催化活性,建立高灵敏度诺如病毒检测方法。方法首先进行纳米酶合成、表征及酶活力测定,然后通过纳米酶与诺如病毒抗体偶联,制备诺如病毒纳米酶-抗体探针,组装成纳米酶试纸条,用诺如病毒纳米酶试纸条检测不同浓度抗原,从灵敏性、特异性等方面对建立的方法进行评价。结果纳米酶表征大小均一,颗粒直径为(100±10)nm,纳米酶比活性9.33 U/mg。研发的纳米酶试纸条对诺如病毒的最低检出浓度为10 ng/ml,比普通胶体金试纸条灵敏度高10倍;与轮状病毒、腺病毒、幽门螺杆菌均无交叉反应。结论纳米酶试纸条检测诺如病毒具有简便、快速、可现场应用的优点,在诺如病毒快速检测中具有实际应用价值。Objective To develop a highly sensitive strip for Norovirus detection by using the catalytic activity of nanozyme.Methods The synthesis,characterization and determination of enzyme activity of nanozyme were performed,and the Norovirus nanozyme-antibody probe was prepared by coupling the nanozyme with Norovirus antibody,which were assembled on the nanozyme strips.Norovirus antigen in different concentration was used to detect by the nanozyme strips.The sensitivity and specificity of the nanozyme strip were evaluated.Results The nanozyme particle was uniform with the diameter of(100±10)nm,the specific activity of the nanozyme was 9.33U/mg.The minimum detection limitation of the established nanozyme strip for Norovirus was 10 ng/ml,which was 10 times sensitive than the traditional colloidal gold strip.The nanozyme strip possessed high specificity for Norovirus,without any cross-reaction with Rotavirus,Adenovirus and Helicobacter pylori.Conclusion The nanozyme strip method had the advantages of simple,rapid and field application,showed application value in the rapid detection of Norovirus.
