摘  要:目的:采用网络药理学的方法探究金银花抗炎的作用机制。方法:通过药理学分析平台TCMSP数据库以口服生物利用度(OB)≥20%和类药性(DL)≥0.1为条件筛选金银花活性成分。并采用Cytoscape3.7软件构建金银花活性成分-预测靶点网络图。利用TTD数据库以“Anti-inflammatory”为关键词搜索抗炎靶点,再使用String数据库构建抗炎相关靶点之间的PPI网络图,并将其与活性成分-预测靶点融合,获得金银花活性成分的抗炎作用靶点。利用DAVID数据库对金银花抗炎作用靶点进行KEGG通路富集分析,以探究金银花抗炎作用机制。结果:获得金银花中具有类药性、口服吸收良好的40个成分,包括槲皮素、芹黄素、山柰酚等。其中抗炎成分主要有金圣草(黄)素、山柰酚、槲皮素、毛地黄黄酮等。金银花抗炎作用的主要靶点有丝裂原活化蛋白激酶14(MAPK14)、表皮生长因子受体(EGFR)、E-选择素(SELE)、受体蛋白酪氨酸激酶erbb-2(ERBB2)等。其中金圣草(黄)素、5-羟基-7-甲氧基-2-(3,4,5-三甲氧基苯基)铬酮、2-(3,4-二甲氧基苯基)-5-羟基-7-甲氧基-铬酮、麦黄酮主要作用于MAPK14;山柰酚、槲皮素作用于SELE;毛地黄黄酮、槲皮素作用于EGFR和ERBB2。KEGG分析得到与金银花抗炎作用有关的通路18条,主要涉及TNF信号通路、MAPK信号通路等。结论:金银花中的活性成分主要通过MAPK14、EGFR、ERBB2、SELE等靶点抑制炎症因子的产生、抑制炎症因子与相应受体结合、阻断炎症反应的启动等,最终发挥抗炎作用。Objective:To explore the anti-inflammatory mechanism of honeysuckle(Lonicera japonica Thunb.)based on network pharmacology.Methods:The pharmacological analysis platform TCMSP database was used to screen the active constituents of honeysuckle under the conditions of oral bioavailability(OB≥20%)and drug-likeness(DL≥0.1).Cytoscape3.7 software was used to construct the network map of the active component-predictive target of honeysuckle.The key words of anti-inflammatory was applied to search the anti-inflammatory targets by using TTD database.String database was used to construct the PPI network diagram among related anti-inflammatory targets.The diagram then was merged with the active ingredient-forecast target in order to obtain the anti-inflammatory targets of the active ingredients of honeysuckle.DAVID database was used to carried out KEGG pathway enrichment analysis of the anti-inflammatory targets of honeysuckle,so as to explore the anti-inflammatory mechanism of the herb.Results:40 medicinal constituents of honeysuckle were obtained,including quercetin,apigenin,kaempferol and other ingredients,among which the main anti-inflammatory ingredients were chrysoeriol,kaempferol,quercetin and luteolin.The main anti-inflammation targets of honeysuckle were mitogen-activated protein kinase 14(MAPK14),epidermal growth factor receptor(EGFR),e-selectin(SELE)and receptor protein erbb-2(ERBB2).Chrysoeriol,5-hydroxyl-7-methoxy-2-(3,4,5-trimethoxyphenyl)chromone,2-(3,4-dimethoxyphenyl)-5-hydroxyl-7-methoxy-chromone and tricin mainly acted on MAPK14;Kaempferol and quercetin acted on SELE;Luteolin and quercetin acted on EGFR and ERBB2.18 pathways related to the anti-inflammatory effect of honeysuckle were identified by KEGG analysis,mainly involving TNF signaling pathway and MAPK signaling pathway.Conclusion:The active components in honeysuckle mainly inhibit the production of inflammatory factors,inhibit the binding of inflammatory factors to corresponding receptors,and block the initiation of inflammatory response through
