摘 要:目的:探讨调脂通脉解毒方对脑缺血损伤大鼠hs-CRP、Lp-PLA2炎症因子表达及脑梗死体积的影响。方法:将雄性SD大鼠随机分为假手术组、模型组、调脂通脉解毒方低剂量组[20 mg/(kg·d)]、调脂通脉解毒方中剂量组[40 mg/(kg·d)]、调脂通脉解毒方高剂量组[60 mg/(kg·d)]及尼莫地平组[15mg/(kg·d)];采用线栓法制备大脑中动脉梗死大鼠模型,在缺血2 h后,拔出线栓实施24 h再灌注,再灌注术后14 d造模成功大鼠灌胃给药,每天1次,假手术组及模型组给予等体积溶液。观察各组hs-CRP、Lp-PLA2的含量变化及脑梗死体积的情况。结果:与假手术组比较,模型组、尼莫地平组、调脂通脉解毒方各组大鼠脑组织hs-CRP、Lp-PLA2含量均显著升高(P<0.01);与模型组比较,调脂通脉解毒方各组、尼莫地平组hs-CRP、Lp-PLA2含量均显著降低(P<0.01)。除假手术组外其余各组均出现脑梗死情况,与模型组比较,调脂通脉方高、中、低剂量组和尼莫地平组均能显著改善脑梗死情况,其中以高剂量组最优。结论:调脂通脉解毒方能显著降低hs-CRP、Lp-PLA2表达,减轻炎症损伤反应,减少脑梗死体积,从而对大鼠缺血再灌注损伤起到保护作用。Objective:To investigate the effects of Tiaozhi Tongmai Jiedu Formula on the inflammatory factors expressions of hs-CRP and Lp-PLA2 and the volume of cerebral infarction in rats with cerebral ischemic-reperfusion injury.Methods:Male SD rats were randomly divided into the sham operation group,the model group,Nimodipine group[15 mg/(kg·d)]and Tiaozhi Tongmai Jiedu groups of low-dose[20 mg/(kg·d)],medium-dose[40 mg/(kg·d)]and high-dose[60 mg/(kg·d)].Rat models of middle cerebral artery infarction were made by suture-occluded method.After 2 hours of ischemia,the suture was removed and reperfusion had been carried out for 24 hours.After successful modeling,the rats were given intragastric administration once a day,the sham operation group and the model group were given equal volume solution.The content changes of hs-CRP and Lp-PLA2,as well as cerebral infarction volume were observed after interventions in the groups.Results:Compared to those in the sham operation group,the contents of hs-CRP and Lp-PLA2 in brain tissue of the rats were significantly increased in the model group,Nimodipine group and Tiaozhi Tongmai Jiedu groups(P<0.01);of which the decreases could been seen after the interventions of Tiaozhi Tongmai Jiedu Formula and Nimodipine when compared to those in the model group(P<0.01);the effect was most significant in Tiaozhi Tongmai Jiedu group of high-dose.Conclusion:Tiaozhi Tongmai Jiedu Formula can significantly reduce the contents of hs-CRP and Lp-PLA2.It can reduce inflammation and cerebral infarction volume,thus protecting rats from ischemia-reperfusion injury.
