摘  要:目的:探讨促红细胞生成素(EPO)对老年性耳聋小鼠血清和耳蜗组织中白细胞介素-6(IL-6)和白细胞介素-17(IL-17)表达的影响。方法:使用SPF级雄性5周龄小鼠48只。其中12只为空白对照组(CK组),36只行老年性耳聋的造模后,分为EPO干预组(EPO组)、生理盐水干预组(NS组)和对照组(C组)。EPO组应用EPO共8周,NS组应用生理盐水8周,对照组不进行任何处理。应用免疫组化法检测各组小鼠耳蜗组织中IL-6和IL-17的表达,应用ELISA法检测各组小鼠血清中IL-6和IL-17的表达,应用实时荧光定量PCR法检测各组耳蜗组织中IL-6 mRNA和IL-17 mRNA的表达水平。结果:免疫组化结果显示:C组和NS组IL-6和IL-17的阳性率明显高于CK组,EPO组IL-6和IL-17的阳性率明显低于C组和NS组。ELISA结果显示:C组和NS组血清中IL-6和IL-17的表达量明显高于CK组,EPO组IL-6和IL-17的表达量明显低于C组和NS组。实时荧光定量PCR结果显示:C组和NS组耳蜗组织中IL-6 mRNA和IL-17 mRNA的表达量明显高于CK组,EPO组IL-6 mRNA和IL-17 mRNA的表达量明显低于C组和NS组。结论:EPO可以减少老年性耳聋小鼠模型血清和耳蜗组织中IL-6和IL-17的表达,减少局部的炎症反应,对延缓病变的进展及促进病变恢复可能有一定价值。Objective:To investigate the effect of erythropoietin on the expression of interleukin-6(IL-6)and interleukin-17(IL-17)in serum and cochlear tissue of senile deafness mice.Methods:48 SPF male 5-week-old mice were used.12 of them were selected as blank control group(CK group),36 of them were selected for senile deafness model and divided into EPO intervention group(EPO group),NS intervention group(NS group)and control group(C group)after modeling.The EPO group was treated with EPO for 8 weeks,the NS group with normal saline for 8 weeks,and the control group without any treatment.The expression of IL-6 and IL-17 in the cochlea tissue of mice was detected by immunohistochemistry.The expression of IL-6 and IL-17 in the serum of mice was detected by ELISA.The expression of IL-6 mRNA and IL-17 mRNA in cochlea tissue of mice was detected by real-time fluorescence quantitative PCR.Results:The results of immunohistochemistry showed that the positive rates of IL-6 and IL-17 in C group and NS group were significantly higher than those in CK group,and the positive rates of IL-6 and IL-17 in EPO group were significantly lower than those in C group and NS group.The results of ELISA showed that the expression of IL-6 and IL-17 in serum of C group and NS group was significantly higher than that of CK group,and the expression of IL-6 and IL-17 in EPO group was significantly lower than that of C group and NS group.The results of real-time fluorescent quantitative PCR showed that the expression of IL-6 mRNA and IL-17 mRNA in the cochlea tissue of C group and NS group was significantly higher than that of CK group,and the expression of IL-6 mRNA and IL-17 mRNA in EPO group was significantly lower than that of C group and NS group.Conclusion:EPO fcan reduce the expression of IL-6 and IL-17 in serum and cochlear tissue of senile deafness mice,reduce the local in-lammatory response,which may be of some value in delaying the progress of pathological changes and promoting the recovery of pathological changes.
