摘  要:目的:研究陈皮对幼龄厌食大鼠的作用及机制。方法:大鼠随机分为对照组、模型组、三个给药组,每组12只。空白对照组常规饲料喂养,模型组用特制饲料喂养;在造模8 d后开始给药,对照组和模型组灌胃生理盐水,给药组分别灌胃10、30、50 g/kg的陈皮水煎剂。通过称重法测定大鼠摄食量、体重和胃排空,采用Mett法测定胃蛋白酶活性,中和滴定法测定胃酸含量,试剂盒测定血浆中胃动素、胃泌素和β-内啡肽,蛋白印迹法检测下丘脑中瘦素和神经肽Y蛋白的表达。结果:与模型组相比,给药组大鼠摄食量和体重显著增加,胃蛋白酶活性显著增强,胃酸分泌量明显增多,血浆中胃动素、胃泌素和β-内啡肽含量显著增多,下丘脑中瘦素明显下调、神经肽Y明显上调,胃残留物显著减少(P<0.01);且效果随着给药量的增加越显著。结论:陈皮呈剂量依赖性改善幼龄大鼠厌食症,主要与增强胃蛋白酶活性,增多胃酸分泌量及血浆中胃动素含量、胃泌素含量、β-内啡肽含量,下调下丘脑中瘦素和上调神经肽Y,促进胃排空有关。Objective:To investigate the effect and mechanism of Pericarpium cirri reticultae on young anorexia rats.Methods:The rats were randomly divided into a control group,a model group,and three drug-administering groups,12 rats each.The blank control group was fed with normal feed,the model group was fed with special feed,and the administration was started 8 days after modeling.The control group and the model group were given normal saline,and the administration group was intragastrically administered with different doses of dried tangerine peel decoction;the food intake,body weight and gastric emptying of each group were determined by weighing method.Pepsin activity was determined by Mett method,gastric acid content was determined by neutralization titration,plasma motilin,gastrin andβ-endorphin were measured by a kit,and the expression of leptin and neuropeptide Y protein in the hypothalamus was detected by Western blotting.Results:Compared with the model group,the food intake and body weight of the rats in the drug-administered group increased significantly,the pepsin activity increased significantly,the gastric acid secretion increased significantly,the levels of motilin,gastrin andβ-endorphin in plasma increased significantly,leptin was significantly down-regulated and neuropeptide Y was up-regulated in the hypothalamus,and the gastric residue was significantly decreased(P<0.01).The effect was more significant with the increase of the dose.Conclusion:Pericarpium cirri reticultae improves anorexia in young rats in a dose-dependent manner.This is mainly related to enhancing pepsin activity,increasing gastric acid secretion,increasing plasma motilin,gastrin andβ-endorphin content,down-regulating leptin,upregulating neuropeptide Y,and promoting gastric emptying.
