摘 要:采用超声联合减压内部沸腾法提取吴茱萸果实中吴茱萸碱与次碱,并利用双波长分光光度法测定其含量。以吴茱萸碱和吴茱萸次碱提取率为指标,通过单因素试验和正交试验考察超声波提取、减压提取对得率的影响,确定最佳提取工艺。其最佳条件为:解吸剂用量6 mL、解吸时间20 min、操作压力0.03 MPa、减压时间6 min时。结果表明,该方法在最优条件下吴茱萸碱提取率为2.34%,吴茱萸次碱提取率为0.81%。与传统炮制法、超声波法和微波法提取相比,用该试验方法提取,所需温度低、试剂少、效率高,是提取吴茱萸碱和吴茱萸次碱较好的一种新方法。Ultrasonic combined decompress inner ebullition was used to extract evodiamine and rutecarpine from the fruit of Evodia rutaecarpa,and the content was determined by dual wavelength spectrophotometry.The Influence of ultrasonic extraction and decompress inner ebullition extraction on the yield of evodiamine and rutecarpine was studied in single factor and orthonormal experiments,and the optimal conditions were determined.The optimal conditions were 6 mL desorption agent,20 min desorption time,0.03 MPa operating pressure,6 min decompression time.The results showed that this method could effectively extract evodiamine and rutecarpine from fruits of Evodia rutaecarpa;under the optimal conditions,extraction yield of evodiamine and rutecarpine were 2.34%and 0.81%respectively.In comparison with the conventional method,such as processing method,ultrasonic method,and microwave method,this new method is a better method to extract evodiamine and rutecarpine in low temperature,less reagents and high efficiency.
