白芷和石菖蒲配伍对大鼠脑缺血损伤的保护作用

摘　　要：目的研究白芷和石菖蒲配伍对大鼠脑缺血损伤的保护作用。方法给大鼠连续灌胃给药7 d后,用线栓法制作永久性大脑中动脉阻塞(MCAO)大鼠脑缺血模型,观察大鼠神经症状、脑梗死范围和脑含水量;利用生化比色法测定脑组织中乳酸(LD)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)及丙二醛(MDA)的含量;用Elisa法测定脑组织中白介素-1β(IL^-1β)、白介素-6(IL-6)、白介素-8(IL-8)和肿瘤坏死因子-α(TNF-α)的含量。结果白芷香豆素组及配伍组大鼠脑梗死范围明显减小,脑含水量明显降低;配伍组对神经缺损症状和梗死范围的改善作用明显优于白芷香豆素组。白芷香豆素和石菖蒲挥发油配伍后可显著提高脑组织中LDH和SOD的含量;白芷香豆素组和配伍组各炎症因子含量均明显降低,但配伍组降低IL^-1β和IL-8含量的作用明显优于白芷香豆素组。结论白芷和石菖蒲配伍通过抗炎和抗氧化发挥抗脑缺血作用,石菖蒲可增强白芷的抗脑缺血作用。Objective To investigate the protective effects of the compatibility of Angelica dahurica and Acorus tatarinowii on cerebral ischemic injury in rats.Methods Permanent middle cerebral artery occlusion(MCAO)rat model was made by using the intraluminal filament method after administration for 7 d.The neurological symptoms,the size of cerebral infarction and the water content of brain tissue were observed.The contents of lactate(LD),lactate dehydrogenase(LDH),superoxide dismutase(SOD)and malondialdehyde(MDA)in brain tissue were determined by biochemical colorimetry,and the contents of interleukin-1β(IL^-1β),interleukin-6(IL-6),interleukin-8(IL-8)and tumor necrosis factor-α(TNF-α)in brain tissue were determined by Elisa method.Results Coumarins group and compatibility group of coumarinsmarins and volatile oils are significantly decreased the cerebral infarct volume and reduced the edema degree in rat brain tissue.The effect of compatibility group of coumarins and volatile oils on the improvement of neurological deficit symptoms and infarct size was better than that of coumarins group.The compatibility of coumarins and volatile oils could significantly increase the activities of SOD and LDH in brain tissue,and the contents of inflammatory factors in coumarin group and compatibility group decreased significantly.Moreover,the effect of combination group on reducing the contents of IL^-1βand IL-8 was significantly better than that of coumarin group.Conclusion The compatibility of Angelica dahurica and Acorus tatarinowii has obvious anti-cerebral ischemia effect,and its neuroprotective mechanisms are mainly related to anti-free radical and anti-inflammatory.Acorus tatarinowii could enhance the anti-cerebral ischemic effect of Angelica dahurica.
