摘  要:目的研究大黄素(EMD)对过氧化氢及叔丁基过氧化氢(t-BHP)分别诱导的L02细胞损伤的保护作用。方法以L02细胞为研究对象,用过氧化氢及t-BHP建立2种L02细胞氧化应激损伤模型。用噻唑蓝(MTT)法检测大黄素对L02细胞活性的影响;用DCFH-DA荧光探针检测大黄素对过氧化氢及t-BHP分别诱导的L02细胞内活性氧(ROS)的水平;用微板比色法检测大黄素对L02细胞中丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)水平的影响。结果过氧化氢及t-BHP均能显著抑制L02细胞活力;中、高浓度给药组(16和160μmol·L^-1)对过氧化氢及t-BHP分别诱导的L02细胞活力下降均有明显的抑制作用(P<0.05);低、中和高浓度给药组(1.6,16和160μmol·L^-1)可抑制过氧化氢及t-BHP分别诱导的L02细胞内ROS、ALT和AST水平的升高(P<0.05)。结论大黄素对过氧化氢及t-BHP分别诱导的L02肝细胞损伤均具有保护作用。Objective To study the protective effect of emodin(EMD)on L02 cells injury induced by hydrogen peroxide and tert-butyl hydrogen peroxide(t-BHP).Methods Taking L02 cells as the research objects,2 oxidative stress injury models of L02 cells were established with hydrogen peroxide and t-BHP.3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT)assay was used to detect the cell activation of L02 cells treated by different concentration emodin.The level of reactive oxygen species(ROS)was detected by DCFH-DA fluorescence probe.Alanine transaminase(ALT)and serum aspartate transaminase(AST)were detected by micro plate and colorimetric method.Results Hydrogen peroxide and t-BHP could significantly inhibit cell viability.Middle and high concentration groups(16 and 160μmol·L^-1)had significant inhibitory effects on the decreased activity of L02 cells induced by hydrogen peroxide and t-BHP,respectively(P<0.05),while the low,middle and high concentration groups(1.6,16 and 160μmol·L^-1)inhibited the intracellular ROS,ALT and AST levels(P<0.05).Conclusion Emodin can protect L02 hepatocyte injury induced by hydrogen peroxide and t-BHP.
