摘　　要：人沉默调节蛋白6(SIRT6)是在哺乳动物中sirtuins家族SIRT1-SIRT7中的一员。2000年,在酵母体内发现沉默信息调节子2(Sir2),实验中发现该蛋白起到延长酵母寿命的作用。SIRT6有多种蛋白酶功能,其中以烟酰胺腺嘌呤二核苷酸(NAD+)依赖的蛋白去乙酰化酶和腺苷二磷酸(ADP)核糖基转移酶活性最为人熟知。SIRT6定位在细胞核内,通过对组蛋白和非组蛋白去乙酰化或对某些蛋白底物行核糖基化,在细胞活动的多个方面,包括转录沉默、细胞凋亡调控、脂肪动员和寿命调控等过程中发挥重要作用。本文仅从其生理功能与凋亡相关性方面作一介绍。Sirtuin(SIRT6)is a member of the sirtuins family SIRT1-SIRT7 in mammals.In 2000,silent information regulator 2(Sir2)was found in yeast.It was found that the protein played an important role in prolonging the longevity of yeast.SIRT6 has many proteases,among which NAD+dependent protein acylase and ADP ribosyltransferase are most well known.SIRT6 is located in the nucleus.By the acylation of histone and non histone or ribonylation of some protein substrates,it plays an important role in the multiple surfaces of cell activity,including transcriptional silence,apoptosis regulation,fat mobilization and life regulation.This article only introduces its physiological function and its correlation with apoptosis.
